Ambroxol In The Lungs Protection From Acute Lung InjuryOBJECTIVES: To explore the mechanism of ambroxol in the protection from acute lung injury (ALI) by studying the change of cytokine mRNA expression during the course of ALI.METHODS: The indexes of test in vivo or in vitro include: Pathological alteration, lung index, arterial partial pressure of oxygen(PaO2), Expressions of TNF-α,IL-10,IL-24 mRNA were determined by RT-PCR in ALI rats given ambroxol as well as avleolar macrophages given 10 mmol/L LPS and 180μmol/L ambroxol cultured in vitro for 6, 12 and 24 hours. RESULTS: 1) The lung index (LI) was increased in ALI rats with an dramatic decrease of PaO2, which can be partially recovered given ambroxol. Massive hemorrhage along with engorgement, edema and infiltration occurred in the lungs of ALI rats. The pathological alteration in ALI rats given ambroxol was less severe. 2) TNF-α,IL-10,IL-24 mRNA expressions were dramatic increased in ALI rats, and partially reduced after treated with ambroxol.3) Macrophages for showed a dramatic increase in the expression of TNF-α,IL-10,IL-24 mRNA in 6 hours exposed to LPS and remain a high level afterwards. The expressions of TNF-α,IL-10,IL-24 mRNA in macrophages exposed to both LPS and ambroxol were less increased than those exposed to LPS solely. CONCLUSIONS: 1) The ALI rat model can be set up by yeast suspension injection to the abdominal cavity. 2) The alveolar macrophage is activated in the inflammatory response of the ALI. 3) The increase of TNF, IL-10 and IL-24 mRNA expression occurred in ALI is possibly due to the activation of macrophage by LPS.４) Ambroxol can partially decreased the expressions of TNF-α,IL-10,IL-24 mRNA in alveolar macrophage induced by LPS.
