Interferon- λ1 (IFN-λ1) is a novel cytokine which was founded in 2002. IFN-λ1 expression was detected at low amounts in libraries from a wide range of tissue types, including blood , brain , lung , overy .pancreas .pituitary , placenta , pro state and testis. IFN-λ1 induce transcription of genes encoding 2' ,5' -OAS and MxA protein and up-regulation of MHC class I antigen expression which mediate antiviral activation . To lookfor a good method to producing IFN-λ1, we designed a project which are carried out through gene cloning and cell engineeringtechniques to provide the problem.Part I Cloning of Human IFN-λ1 GeneObjective: To study a new method of obtaining IFN-λ1 gene. Method The total RNA was isolated from A549 cells whichinduced by Vesicular stomatits Virus (VSV) ,then the enoughamounts cDNA of IFN-λ1 was obtained by using Reverse transcribed polymerase chain reaction (RT-PCR) with a pair ofspecific primers. Results The amplified product by RT-PCR wasthe full-length sequence encoding human IFN-λ1 (603) by electrophoresis in agarose gels and DNA sequencing .Conclusion The clone containing the full length sequence encoding humanIFN-λ1 is obtained successfully.
