| Objective: To observe the proliferation and differentiation properties of mouse skeletal muscle satellite cell and the effect of IGF-1 on it,which is cultured and purified in vitro,and make preparation for satellite cell transplanting.Methods: the cells are isolated by two-step enzyme digestion and purified by the Percoll density centrifugation,the proliferation and differentiation propertiesof which being observed.To study the effect of IGF-1 on satellite cell with MTT method and obtain adequate single satellite cell by frequent passaging.Conclusion: Plenty and purified satellite cells are obtained with these methods.IGF-1 could promote the proliferation and differentiation of mouse skeletal muscle satellite cell. Adequate single satellite cell could be obtained by frequent passaging method and the preparation are ready for for satellite cell transplanting. |
