The Study On The Expression And Correlativity Of P73,p16～(Ink4a),cyclinD1,p27～(Kip1),CDK4 And Ki67 In Gastric Carcinoma

Objective: To evaluate the relationship between the expression of p73, p16Ink4a, cyclinD1,p27Kip1, CDK4 and ki67 in gastric carcinoma and the clinicopathological parameters and to investigate the correlativity of the genes. To analysis the significance of p73, p16Ink4a, cyclinD1, p27kip1, CDK4, ki67 in the occurrence and development of gastric carcinoma in order to provide the theroretical basis for the clinical diagnosis and genetic therapy of gastric carcinoma .Methods: The SP immunohistochemical method was used to detect the expression of p73, p16Ink4a, cyclinD1, p27kip1, CDK4, ki67 in gastric carcinoma and normal gastric mucosa. Results was analysed by SAS version 8.0.Results: 1 .Overexpression of p73 protein was found in 26/68(38.24%) of gastric carcinoma. The positive expression rate of p73 protein in the gastric carcinoma was significantly higher than in the gastric normal tissues 1/20(5%)(P<0.01). No significant relationship was observed among p73 protein expression and age, sex,pathological classification and depth of invasion of the cancer (P>0.05). Significant relationship was observed among p73 protein expression and histological grade, lymph node metastasis and clinical stages of the cancer. The positive rates of p73 protein expression in the cases of histological grade I were significantly lower than in the cases of histological grade â…¡,â…¢ (P<0.01). No significant difference was observed among p73 protein expression and histological grade â…¡, â…¢ (P>0.05). The positive rates of p73 in the cases with lymph node metastasis were significantly higher than those in the cases without lymph node metastasis(P<0.01). The positive rates of p73 in the cases of clinical stages â… -â…¡ were significantly higher than those in the cases of clinical stages â…¢-â…£(P<0.01).2. The positive expression rate of p16Ink4a and p27kip1 protein in gastric carcinoma was 57.35% (39/68) and 44.12% (30/68) respectively, whereas 85.00% (17/20) and 75% (15/20) found in the gastric normal tissues respectively. The positive expression rate of p16Ink4a, and p27kip1 protein in the gastric normal tissues was significantly higher than in the gastric carcinoma (P<0.01). Significant relationship was observed among p16Ink4a and p27kip1 protein expression and differentiated grades, lymph node metastasis and clinical stages of the cancer. The positive rates of pl6Ink4a and p27kip1 protein expression in the cases with high-moderate differentiated grades were significantly higher in the cases with low differentiated grades (P<0.01). The positive rates of p16Ink4a and p27kip1 in the cases with lymph node metastasis were significantly lower than those in the cases without lymph node metastasis(P<0.01). The positive rates of p16Ink4a and p27kip1 in the cases of clinical stages â… -â…¡ were significantly higher than those in the cases of clinical stages â…¢-â…£(P<0.01 ).3. The positive expression rate of cyclinDl and CDK4 protein was 0% and 20% in gastric normal tissues and 73.53% and 61 .76% in the gastric cancer. Difference wassignificant between those in gastric normal tissues and those in the gastric cancer (P<0.01). Significant correlationship was observed among cyclinD1 and CDK4 protein expression and histological grade, lymph node metastasis and clinical stages of the cancer. The positive rates of cyclinDI and CDK4 protein expression in the cases with high-moderate differentiated grades were significantly lower than those in the cases low differentiated grades (P<0.01). The positive rates of cyclin D1 and CDK4 in the cases with lymph node metastasis were significantly higher than those in the cases without lymph node metastasis(P<0.01). No significant relationship was observed among the positive rates of cyclinDI in the cases of clinical stages â… -â…¡ and those in the cases of clinical stages â…¢-â…£(P0.05).
