AIM: By means of Differential Display analyzed the effect of estradiol by concentration of physiological on gene expression of the bone marrow macrophage.Methods: The bone marrow macrophages (BMMs) were separated from three week-aged male mice. The cells were treated with estradiol by 10nM melted in ethanol or ethanol by 0.04%. Through the method of differential display we selected fragments expressed differentially and cloned them into T vector, then sequenced. The obtained sequences were searched against Genbank and then analyzed with BLAST analysis system. Primers were designed according to known genes and then checked by RT-PCR.Results: A new gene and a fragment of ankyrin-1 were found in estradiol treated and control group. The two pairs of primers were synthesized and responded products were amplified in treated group and control group.Conclusion: The study demonstrated that the expression of genes between test group and control group was different, which was related to estradiol and will be helpful to understanding the effect of estradiol on macrophage.
