It is the key of modernization of traditional Chinese medicine (TCM) to study the mechanism of TCM.However,the complex compounds and multitargets of TCM make it difficult to study the mechanism.In this paper, we investigated the effect of combined compounds (CC) of SI-WU-decoction (SWD) on the proliferation of HFCL cells and the blood enriching effect on the Balb/c mice irradiated by 60Co Y .Combinding the results of differential expressed genes detected by oligonucleotide microarrays, we tried to elucidate the relationship between the compounds and mechanism of SWD.1.The CC1 and CC2 of SWD could enhance the proliferation of HFCL cells. Hematopoietic-related gene expression of HFCL cells showed that the genes of IL-2R, NF- K. ^ TPO were up-regulated after HFCL cells exposed to CC1 12h. It also showed that the genes of NF- K b,G-CSF-R, IL-13,VEGF,IL-5,GM-CSFR-a were up-regulated after HFCL cells exposed to CC2 12h. Combinding the gene expression results of HFCL cells exposed to SWD,we found that the mechanism of SWD correlated with the specific compounds - founctional compounds - of SWD. Combinding the gene expression results of HFCL cells exposed toCCl,CC2 and SWD.We found that the mechanism of SWD correlated with not only the specific compounds -founctional compounds-of SWD but also with their relative proportion. It was found that fructose positively correlated to saccharose, the paeoniflorin to tetramethylpyrazine.in enhancing the proliferation of HFCL cells.2. The CC1 and CC2 of SWD could increased leucocyte level and the colony forming units(CFU GM,BFU E,CFU E,CFU mix) of the blood deficiency mice.The result showed that compounds of CC1 was only a part of founctional compounds ofSWD. So the effect of CC1 on the proliferation of CPU E, CFU mix was less than that of SWD.The gene expression results of marrow also showed that the blood enriching mechanism of SWD related with not only the specific compounds - founctional compounds-of SWD but also with their relative proportion. |
