Objectives To investigate the expression of PTEN(phosphatase and tension homolo-gy deleted on chromosome ten)mRNA in gastric carcinomas and their adjacent tissues as well as to appreciate its clinical significance.Methods The expression of PTEN mRNA in 45 cases of gastric carcinomas and its adjacent tissues,was detected by reverse transcription- polymerase chain reaction? the relative mean expression of PTEN mRNA in these samples was detected by semi-quantitative RT-PCR.Results Of the 45 gastric carcinomas, the respectively positive expression of PTEN mRNA was exon 1-8 39(86. 67%) ;exon 5-8 37(82. 22%) ;exon 5-9 39(86. 67%) ; However, the positive expression of PTEN mRNA was found in all of the 45 gastric carcinomas adjacent tissues. Of the 45 gastric carcinomas, the relative mean expression of PTEN mRNA to -Actin was exon exon 1-80.71 0.21,5-8 0.90 0. 11, exon 5-9 0. 83 0. 15. Of the 45 gastric carcinomas adjacent tissues, the relative mean expression was exon 1-8 1. 29 0. 18,exon 5-8 1. 51 0. 27, exon 5-9 1. 51 0. 27. The expression of PTEN mRNA did not show correlation with such factors as sex,age, but had much to do with the clinical staging and histological grading.Conclusion The expression level of PTEN mRNA in gastric carcinomas was sharply reduced compared with their adjacent tissues . Which suggests that PTEN mRNA inactiva-tion plays an important role in the genesis and development of gastric carcinomas. The expression of PTEN mRNA was expected to be the diagnostic criteria of gastric carcinomas. |
