ObjectiveTyrosine protein kinase (TPK) is a kind of protein kinase that could catalyze protein tyrosine phosphorylation, exiting extensively in the human bodies. By catalyzing substrate protein tyrosine phosphorylation, tyrosine protein kinase participated in cell signal transduction. The protein phosphorylation and dephos-phorylation are key to cell signal transduction, between cell growth, differentiation, transfer and tyrosine phosphorylation status are closely related. Phosphoty-rosine(P - Tyr) is product of tyrosine protein kinase catalysis, phosphotyrosine examination is important to study the function and activity of TPK and plays an important role to tumor occurrence, growth, prevention. The purpose of this study was to evaluate the change of tyrosine phosphorylation status in endometrial carcinoma, atypical hyperplasia and normal endometrium, and to explore the significance of the activity of tyrosine protein kinase in the occurrence of endometrial carcinoma.MethodsClinical specimens were obtained from the Department of Pathology, The Second Hospital of China Medical University,for the period May 2000 to March 2003. Patients'ages ranged from 43 to 70 years. Using SP immunohistochemisty method, the expression of phosphotyrosine(P -Tyr) in the specimens of 40 cases with endometrial carcinoma, 17 cases with atypical hyperplasia and 28 cases with normal endometrium were examined, thus getting the activity of tyrosine protein kinase in the three groups.ResultsThe positive immunostaining rate of P - Tyr in cases with endometrial carcinoma, atypical hyperplasia and normal endometrium were 75% (30/40), 47. 06% (8/17) ,and 14.29% (4/28) respectively,there was significant difference between cancer tissues and normal endometrium or atypical hyperplasia (P <0.05) ,the activity of tyrosine protein kinase in endometrial carcinoma was much higher than that in atypical hyperplasia and normal endometrium. In the 40 cases of endometrial carcinoma, the expression of P - Tyr in moderately - poorly differentiated was significantly higher than that in well differentiated, the positive immunostaining rate were 90.48% (19/21) and 57. 89% (11/19) respectively, there was significant difference(x =4. 043,P <0. 05) . but not correlated with age(x2 =0.711,P>0.05), clinical stage(x2 =0. 159,P>0.05) ,muscles invasion ( x2 = 0. 853 , P > 0. 05 ) , lymphatic gland transfer ( x2 = 0. 686, P > 0.05).Conclusion1. The abnormal activity of tyrosine protein kinase was the cause with endometrium abnormal growth.2. The activity of tyrosine protein kinase is correlated with histological grade,but not with age, clinical stage,muscles invasion,lymphatic gland transfer.
