| To explore anti-aging effect and mechanism of Lipoteichoic acid of Bifidobacterium.Male Kunming mice were divided into different groups at random. These mice were induced aging by subcutaneous injection of D-galactose on the back and LTA was simultaneously administered to them daily. After 40 days, mice were killed and these indexes were measured such as increasing curve of body weight, organ index, the morphology of kidney, liver and thymus by HE stain, the activities of SOD and NOS, the levels of NO and MDA in brain, the concentration of IL-2 in blood by ELISA , and the production of p16 and c-fos in kidney, liver and thymus by immunohistochemistry , DNA damage of lymphocyte in spleen by single cell gel electrophoresis and nucleus chromosomal aberration in bone marrow by Giemsa stain.There were significant differences in body weight , the brain index, the kidney index ,the thymus index, the spleen index (P<0.05 P<0.01, P<0.01, P<0.01, P<0.05 ) between LTA group and aging model group; LTA of bifidobacterium can noticeably increase the activities of SOD and NOS, the level of NO (P<0.05, P<0.01, P<0.01), decrease the level of MDA in brain (P<0.01) and enhance the concentration of serum IL-2 (P<0.01) . Besides, LTA of bifidobacterium may remarkably reduce the appearance rate of comet and the length grade of DNA between the LTA group and the aging model group (P<0.05) . Moreover, the mice treated by LTA had fewer micronucleus frequency than the aging model mice (P<0.01) .The results above suggested that LTA of bifidobacterium has anti-aging effect and the mechanisms may be as follows: firstly, it can obviously improve the ability of anti-oxidation in brain. Secondly, it can decrease pi6 production and meanwhile enhance c-fos production in liver, kidney and thymus. Besides, it can strengthen the immunological function by decreasing DNA damage of lymphocytes in spleen and inhibiting nucleus chromosomal aberration in bone marrow. Simply put, many mechanisms about bifidobacteriun anti-aging were revealed. |
