Objective: To investigate whether the conditioned media of hypothalamus-pituitary(HP) and minimally modified low density lipoprotein(mmLDL) influences the development and rupture of atheromatous plaque by using conditioned media of cultured hypothalamus-pituitary cells to culture macrophages induced by mmLDL , then the content of tumor necrosis factorα (TNFα) and interleukin-1β( IL-1β) was detected. Methods: The hypothalamic and pituitary tissues from freshly aborted 5-month human fetus were taken for cellular culture and the conditioned media were collected. The macrophages of HP group were cultured with the conditioned media, the HP- group with ordinary media. The time required was 24h. After induced by mmLDL for 20h, all the macrophages of the experimental and control groups were cultured in serum-free RPMI1640 media for 36h. Then the supernatants wer collected respectively for detecting the content of TNFαand IL-1β with radio-immunity method. Results: It was shown that: (1) The changes of TNFαand IL-1β production by macrophages induced by mmLDL showed up-regulation compared to control (P<0.05); (2)The conditioned media of hypothalamus-pituitary had downregulation effect on the content of TNFα(P<0.05) and upregulation effect on the IL-1β expression (P<0.05). Conclusion: It can be concluded from these results that when a pathogenetic factor of AS , such as mmLDL, induces macrophages, the responsive secretion of macrophages including TNFα and IL-1β. Simultaneously the pathogentic factor stimulates the responsive secretion of the neuroendocrinesystem that down-regulating the content of TNFαand up-regulating the release of IL-1β. The hypothalamus-pituitary and mmLDL may play an key role in the development of the development and rupture of atheromatous plaque. The results also provide a theoretical base for the central pathogenetic mechanisms of AS .
