Objective: To study the relationship between the mutation of PTEN and the endometrioid carcinoma. Methods: The mutations of exon 1-9 in the PTEN gene of 32 endometrioid adenocarcinomas tissues and 10 endometrioid hyperplasiases tissues were examined by polymerase chain reaction and DNA sequence analysis. Results: 43 mutations were detected in the endometrioid adenocarcinomas.3 mutations were detected in exon 2. 4 mutations were detected in exon 4. 18 mutations were detected in exon 5. 16 mutations were detected in exon 6. 1 mutations were detected in exon 7. The mutation was not detected in exon 1,3,8 and 9. Of 43 mutations 19 were frameshift. 17 were missense, 7 were nonsense. The mutation rate(22/32)68.75% in endometrioid adenocarcinomas was higher than that of endometrioid hyperplasiases tissues(0), p<0.01. The mutation frequence in endometrioid adenocarcinoma without papillary cases (21/27) 77.8% was higher than that of in endometriod adenoncarcinoma with papillary cases (1/5) 20%, p<0.05. The mutation frequency in well-differentiated cases (15/25) 60% had no significant difference compared with that of poor-differentiated cases (7/7) 100%,p>0.05. The mutation rate in <1/2 depth of myometrial invasion cases (14/20) 70% had no significant difference compared withthat of in >1/2 depth of myometrial invasion cases (7/7) 100%,p>0.05.The mutation rate in stage I-II cases (20/29) 68.9% had no significant difference compared with that of stage III cases (2/3) 66.67%, p>0.05. It was that PTEN mutations were not confined to pathological grade and depth of myometrial invasion and stage. Conclusion: The mutation of PTEN is an important molecular event in endometrioid carcinoma. The majority of mutations accured in exon 5. Most of mutation predicted truncated PTEN protein products by small deletions or insertions. PTEN was a target gene in endometrioid adenocarcinoma. Molecular basis of endometrioid adenocarcinoma without papillary was not different with that of endometrioid adenocarcinoma with papillary.
