Objective: Study on the therapeutic role of mixture Dan Shen, Da Huang(DD) and arginine, taurine to acute experimental hepatitis by CCL4 and the therapatic effect of DD on chronic experimental hepatitis and it's mechanisms.Methods: Part I All the animals were divided into four groups at random, called separately normal group, acute experimental group, DD group, DD+arginine+taurine group with 6 rats in each group. The three groups except the normal group were given subcutaneous injection of carbon tetrachloride(CCL4) in the 1st and 5th day. The DD and DD+arginine+taurine groups were given DD and mixture of DD+ arginine+taurine. The rats were killed in the 8th day. Blood was taken from abdominal aorta carefully to test the plasma ET, ALT, EL-2, IFN-r, IL-4. At the same time, observed the vary of liver histology.Part II The rats were divided into 3 groups at random, called seperatly normal group, chronic experimental hepatitis group and DD group with 10 animals each. The model was made by subcutaneous injections of CCL4and dietetic factors till the end of 4th week. Blood was taken from abdominal aorta carefully to test the plasma ET, ALT, IL-2, IFN-r, IL-4. At the same time, observed the vary of liver histology. Results: (l)The level of ET and ALT increased significantly in the acute experiment (p<0.01) comparing with normal group. IL-2, IFN- Y were lower than normal group (p<0.05), while IL-4 was higher(p<0.05).The similar results were achieved in chronic experiment.(2)Compared to acute hepatitis group, the level of ET in DD+arginine+taurine group decreased apparently whose effect was not so good as DD group alone. At the same time, the decrease of the level of ALT by DD+arginine+taurine was more apparent than DD alone argtnine and taurine function as cytoprotective factor. IL-4 decreased, while IFN- Y ,IL-2 increased though IFN-r was no evident discrepancy statistically. Conclusions: The increase of ET in acute experimental hepatitis caused by CCL4 may lead to the disorder of cellular immune function which was characterized by the increase of IL-4 and decrease of IL-2 and IFN-r . The disorder also reflected the debalane of Th/Th2. At the same time the hepatocytes were damaged significantly with the characteristic of increase of ALT. After the therapeutics by DD , the level of ET was decreased significantly and the ratio of Th,/Th2 was adjusted and so the ruin ofhepatocyte was alleviated. The similar result was also achieved in chronic experiment. DD+arginine+taurine enhanced the cellular immune function and weaken the ruin of liver because it functions as cellular immunomodulator as well as cytoprotective factor.On the whole all these experimental results may tell what measures should be taken in order to attain satisfied effects.
