Objective: To investigate the anesthetic site and mechanism of isofluraneby evaluating the effect of C-FOS protein repressing in spinal cord of rats whose left femoris bicep was received electrical stimulation. Methods: A rat inhaled isoflurane through a mask after being anesthetized, then its cervical was dissected and a catheter was inserted into the tracheal. Animal ventilator controlled the rat's breath. Thirty-two rats were randomly divided into four groups : group 6F, group 9F, group 11F and group 14F and correspondently inhaled isoflurane in 0.6MAC, 0.9 MAC, 1.1MAC and 1.4 MAC. After having balanced the isoflurane density of alveolus at the end of the expiration. the left femoris bicep received the electrical injured stimulation. One hour later, the rat was perfused with 4%poly-formalindehyde and 0.1M sodium phosphate by aorta. The spinal cord in L4.6 was removed and treated especially to be paraffmed-inbeded and dissected. Sections weret then immunostained for protein using the immunohistochemistry method (ABC). We made a count of all FOS-like-immunoreaction neurons (FLIN) in each specific lamina region on the ispilateral side of electrilic stimulation, classified as follows; superficial layer of dorsal horn (lamina I and â…¡). nucleus proprius (lamina â…¢ and â…£). neck of the dorsalhorn ( lamina â…¤and â…¥). and ventral gray ( lamina â…¥,â…§, â…¨ and â…©). The lamina â… , â…¡, â…¢,â…£,â…¤ and â…¥ were defined as the dorsal horn, and the lamina â…¦ and â…© was defined as the ventral horn. The number of FLIN in the dorsal horn and ventral horn among four groups were compared using one-way analysis of variance and Scheffe's multiple comparison test. Result: The FLIN in lamina â… ,â…¡,â…¤, â…¥ and lamina â…¢,IV of the ipsilateral horn stimulated was observed. In the of ventral gary , the number of FLIN in 14F group significantly decreased in compared with group 6F, group 9F and group 11F(P<0.01). while the difference was not significant between group 6F. group 9F and group 11F.. The number of FLIN in the gary of dorsal horn decreased, depending on the increasing density of isoflurane inhaled and there were the negative correlation between the number of FLI and the density of isoflurane in lamina â… , â…¡,â…¤,â…¥, however, it was not significant between group 6F and group 9F (P>0.05) . Conclusions: 1. The dorsal horn and the ventral horn are probabi one of the anesthesia sites of isoflurane to inhibit the movement reaction after the injured stimulation. especially, the ventral horn plays an important role. 2. Inhaling isoflurane in 1.4MAC can completely abolish the movement reaction after the injured stimulation and lead to the obvious muscle relaxation.
