Subject : Comeal Endotheliitis is a comeal pathological changes which manifestations are endothelial turbidity and edema, thick folds on descemet' s membrane, stromal edema , and keratic precipitates scattering behind the lesion. All most clinical impression diagnosis depend on the history and typical pathological changes of cornea. With the rising incidences of comal endotheliitis, diagnosis of the pathogeny has become increasingly important. Polymerase Chain Reaction(PCR)technology, a highly sensitive and specific method, has been used in a variety of biomedical research studies. The purpose of this reseach is to evaluate the signification of Polymerase Chain Reaction(PCR) technology on etiologic diagnosing corneal endotheliitis and directing treatmeat of it.Material and Method: The study group included 12 cases(12eyes) suffering from comeal eddotheliitis(impression diagnosis) . The control group included 15cases(15eyes) suffering from cataract. The aqueous humors are collected using a syringe on both groups. The means is to specialy proliferate HSV- I DNA and VZV DNA in aqueous humors and detect the PCR products by Agarose gel electrophoresis. These norms are analysised accordingly: positive ratio of HSV- I DNA in aqueous humor of endotheliitis and the control group; positive ratio of VZV DNA in aqueous humor of eddotheliitis and the control group; positive ratio of HSV-I DNA and VZV DNA in aqueous humor of eddotheliitis. Exact probabilities in 2X2 table was used to analyze data. Statistical significance was considered at P<0.05.Results: 41.67% of Seyes (5 cases) were positive for HSV- I DNA in humor of the endotheliitis, but no one was positive for HSV- I DNA in the humor of control group, there was significant difference between the endotheliitis and control group(P<0.05); nocases were positive for VZV DNA either in humor of the endotheliitis or in the humor of control group; there was significant difference between the positive ratio of HSV- I DNA and VZV DNA in aqueous humors of endotheliitis(P<0.05). Conclusion:1. HSV-I is the primary virus which leadings to cornea! endotheliitis.2. By using the PCR to detect HSV-I DNA in humor of the comeal endotheliitis is rapid and simple for etiologic diagnosis and is convient for clinical treatment.3. The conclusion is limited to diagnosis of HSV and VZV. It is necessary to do further for the other pathogeny.
