Objective: To investigate the association of tumor ncrosis factor alpha and tumor ncrosis factor-alpha-converting enzyme gene with diabetic cardiomyopathy .Methods: (1)Male Sprague-dawley rats were fed normal chow(n=5), or high-fat diet (triglyceride(37. 5%, w/w)and cholesterol(1%, w/w)) for one week and then injected intraperitoneally with streptozotocin (STZ, 25 mg/kg) (n=22) and once again after one week and kept on same diet . (2)Oral tolerance glucose test were measured after 4,8 weeks post-STZ, while fasting insulin and triglyceride and total cholesterol were determined after 6 weeks post-STZ. The weight of rats were measured every week during experiment. (3) dissected myocardium was observed under electron microscope. (4) The tumor ncrosis factor alpha and tumor ncrosis factor-a-converting enzyme mRNA expression level in myocardial tissue of normal chow rats and high-fat/STZ rats with diabetic myocardiopathy were assayed by semiquantitative reverse transcription- polymerase chain reaction.Result: (1)The level of blood glucose of oral glucose tolerance test was raised more significantly in the model group than in the control group in 6 weeks(P0. 05).(2) Diabetic cardiomyopathy was verified by the special ultrastructural change of myocardium.(3) Our data suggested that tumor ncrosis factor alpha and tumor ncrosis factor-a -converting enzyme were up-regulation in diabetic myocardio-pathy(0.437 0.078 VS 0.286 0.061 , P
