| ObjectiveAt present, combined chemotherapy is the common modality of the treatment of hematopoietic and lymphoid malignancies. The chemotherapy drugs have stronger side effect, such as the bone marrow suppress that might result in stoping chemotherapy . Primary and secondary drug resistances often has failure in chemotherapy .Therefore, looking for an anti-tumor drug of high efficiency and low toxin is one of the key research direction of anti-tumor management . Malignant tumors being treated with the Chinese medicinal herbs have a long history in our country. In recent years, the mechanisms of anti-tumor effect of the Chinese medicinal herbs have been widely studied, more and more monomer extracted from the Chinese medicinal herbs and plants have been used in tumor treatment. The Chinese medicinal herbs not only have the cytotoxic effect on tumor cell of hematopoietic and lymphoid tissues, but also can improve the body's immunological function, decrease the chemotherapeutic damage to human body and reverse the drug resistances.The Chinese medicinal herbs are less side effects, lower toxic , aboundant in resources and cheap in prices and have the advantages of being used in a long term of anti-tumor over other common chemotherapeutic drugs. Therefore, hematopoietic and lymphoid malignancies treated with Chinese medicinal herbs, natural drugs and effective compounds extracted from them alone or together with chemotherapy have been paid to increasingly more and more attention. However, currently, besides arsenic and elemene, there are not too much effective compounds in the Chinese medicinal herbs used in clinic. It is still necessary to find highly effective, low adverse drug to treat hematopoietic and lymphoid malignancies. Prunella vulgaris L is a traditional Chinese medic.inal herb and has the detoxifcation and detumescence effect. The use of Prunella vulgaris L has a long history in treating malignent tumors. The vitro experiments showed that it can induce the apoptosis of SGC-7910 carcinomatous cells, and the reacting target point is probably between G1 and G2 phase. The vivo experiments of suppressing tumors also indicate that ursolic acid and its derivative containing in Prunella vulgaris L has distinctive cytocity to P388, L1210 and A549 cells of human lung cancer. In current research , Prunella vulgaris L is investigated on the influence of proliferation rates of the three target cells-Erythroleukemia cell line K562, B-lymphoma leukemia cell line Raji and T-lymphoma leukemia cell line Jurkat and on the influence of K562 apoptosis with cell biologic methods in order to offer experimental data for the treatment of hematopoietic and lymphoid malignancies with Prunella vulgaris L injection in clinic. Methods1. Effect of Prunella vulgaris L injection on inhibition ratio of cell growth of K562 cells, Raji cells and Jurkat cells were tested by MTT assay. The research was divided into 3 groups : (1) experimental group, different concentration injection of Prunella vugaris L was administed to culturedtumor cell ; (2) positive control :different concentration of Adriamycin was administed to cultured tumor cell ;(3) control group, the same volume of saline as the former was administed to cultured tumor cell. Every drug concentration which was administed to kill respectively K562 cells, Raji cells and Jurkat cells was set for 4 paralled poles. After 48 hours, the ratio of cell growth was tested and IC50 was figured out. In the meantime, the growth curve line of K562 cells, Raji cells and Jurkat cells which was treated by two different concentration injection of Prunella vulgaris L(50mg/L, 100mg/L) in MTT assay were drawn. Control group was set(the same volume of saline). The above experiments were repeated 3 times respectively, the average value was figured out.2.
