Aim To compare the immune protective effect of three antigens of Trichinella spiralis adult worm on mice .Methods Adult worm of T.spiralis and their medium were collected from the infected rats intestine ,then the adult worm somatic antigen -. adult worm excretory-secretory antigen and adult worm surface antigen were prepared in order to immune the mice. Each mouse was immunized 3 times at 1 week interval by intraperitoneal injection with one of three above antigens lOOMg/O.lml respectively in equal volume of Freund's adjuvant . On day 7 after the final injection, each mouse was challenged orally with 200 infective larvae of T.spiralis . Intestinal adult worms and musle larvae were examined on 7 day and 30 postchallenge respectively. Moreover, the level of serum IgG to antigen of T.spiralis musle larvae was detected by ELISA on 30 days postchallenge. Blood absolute eosinophil level was counted on 7 day and 30 day postchallenge.Results 1. Intestinal adult worm were reduced 84.48%, 89.98%, 85.16% in adult worm somatic antigen adult worm excretory-secretory antigen and adult worm surface antigen groups respectively. Muscle larvae were reduced 69.82%, 78.80?(K 73.94% respectively.2. The antibody titer in all immunized groups increased significantly, the GMRT values of somatic , excretory-secretory and surface antigen groups were 3198.89,3674.5K2784.83 respectively and were 6.96,7.99,6.06 timeshigher than that of unimmunized group(459.19). 3.Periphery blood EOS enhanced since 7 day postchallenge, blood EOS of all geoups increased greatly .The number of EOS in somatic antigen group was 680/mm3 and that of excretory-secretory antigen and surface antigen group were respectively 695/ mm3 , 6151 mm3 .And all of them were significantly higher than that of unimmunized group (270/mm3) (P<0.01).On 30 day postchallenge, EOS of somatic antigen, excretory-secretory antigen and surface antigen group were 940/ mm3,965/ mm3,910/ mm3 respectively, all of them were significantly higher than that of unimmunized group (525/ mm3)(p<0.01) .Conclusion The results show that T.spiralis adult worm antigen can induce strong resistance of host to a subsequent challenge infection and activate specific humoral and cellular immunity .Among these antigens, excretory-secretory antigen is more immunogenic.
