| Objective: To investigate the roles of Activated Nuclear factor- K aapB (NF-KB) p65mRNA, Interleukin-1 β (IL-1 β )mRNA and Interleukin-4 (IL-4)mRNA in the pathogenesis of Mesangial Proliferative glomerulonephritis ,and probe the potential theraptatic way in renal inflammatory. Methods: Using IN Situ Hybridization (ISH) to detection of Activated Nuclear factor- K aapB (NF-KB) p65mRNA, Interleukin-1 β (IL-1)mRNA, Interleukin-4(IL-4) mRNA in biopsy specimens of kidney from patients with Mesangial proliferation glomerulonephritis(MsPGN) (n=41) and in Structurally normal kidneys (n=9). Result: 1. Weak staining of Activated NF-KBp65 , IL-1 β in glomerulus of normal kidney(11.1%,33.3%), Interleukin-4 positive cells were not found in the normal control kidney tissues,the positive rates of three-phase detecting index are higher in glomerulus of MsPGN than in normal kidney.(P<0.05); 2. The positive rate of Activated NF- KBp65mRNA is showed a positive correlation with IL-1 β mRNA (r=0.3836, p<0.05),and also positive correlation with IL-1β mRNA and IL-4mRNA(r=0.4771,p<0.05). 3. A correlation was found between the positive rate of actived NF-KBp65mRNA, II-1β mRNA, IL-4mRNA with the degree of Mesangial proliferation , But no statistic significance. 4.There are not certain regularity in the tubular cells of normal kidney and MsPGN. Conclusion: 1. Activated NF-KBp65mRNA , IL-1β mRNA and IL-4mRNA are associated with the development of MsPGN. 2. It suggests that Activated NF-KBp65 may play an important regulatory effects on the expression of IL-1 β .3 The reactive of Th2 may have relation with the pathogenesis of MsPGN 4 The inhibitor or antagonist of these cytokines may be a new way in treatment of renal inflammatory disorder. |
