Background:The effectiveness of hyperthermia as a treatment modality for magligelant tumors continues to gain popularity in the medical community. This method of treatment, usually palliative, consists of some combination of surgery, radiation, and chemotherapy. Under the treatment background of osteosarcoma, the hyperthermia is used as a kind of effectively of assistance treatment means, already for majority scholar accepts. Hyperthermia, combine with radiotherapy and chemotherapy, already made the treatment result of many kinds tumor a big exaltation. During the past decades, limb salgery has become a standard procedure to treat osteosarcoma and other malignant bone tumors, but there are not possible in nearly 20% to 30% of patients because of tumor size, disease extend, and involvement of the neurovascular structures. Improve treatment result for this part of patients is the key to increase osteosarcoma limb salvage rate and 3 years survive rate.The traditional hyperthermia methods, Radio frequency (RF), ultrasound (US) and microwave (MW), when they transmit in body tissue, echo come into being in the bone surface because of the different medium density between bone and soft tissue. And this reflects to be a certain cause over hot, which may bring hot damage to the main neurovascular band of limb. And there are other limitation for traditional hyperthermia methods, for example, asymmetry of temperature and overheated of fat, etc.Gordon put forward the concept of intrecellar hyperthermia in 1979, and expected with alternating magnetic field induced magnetic nanoparticalsproduces the heat, attaining to the purpose of the even consistent heating in tumor. So we want to use their method to achieve the goal of osteosarcoma hyperthermia pro- or post-operation.This experiment is a trial research about alter magnetic field induce magnetism nanoparticle Fe03 to produce heat, and its hyperthermia effect in vitro and animal model. The purpose is to grope into the magnetic field strength, nanopartical concentration that is fit for osteosarcoma, and the possibility of intracellar hyperthermia, and lays a foundation for its clinical application. Purpose:Through the research of alerting magnetic field induced Fe3O4 nanopartical hyperthermia effect- to osteosarcoma line SOSP-9607, discuss its possibility and potential value of clinical application. Method:1. The cell line SOSP-9607 was cultivated with magnetic Fe3O4 nanopartical RPMI1640 culture-medium, which contain 0.3% Fe3O4 nanopartical in the ratio mass to volume and 10% fetal calf serum in the ratio volume to volume. To determine particle uptake and distribution throughout the cytoplasm, into phagosomes or lysosomes, we observed it with light microscopy, transmission electron microscopy and irons dye in 0.5h, 1h, 2h, 4h, 8h, 16h, 24h, 48h, 72h, 96h, and 120h.2. Survey the temperature change of different Fe3O4 namopartical density RPMI culture-medium in different drive voltage. And the magnetism strength degree at different drive voltage has been measured.3. The freeze-keep pipe with SOSP 9607 osteosarcoma cells cultured in 0.3% Fe3O4 nanometer material culture medium or the tumor cells had cultured in 0.3%Fe3O4 culture medium for 48hours. The duration of irradiation was took 40minutes. Detected the cell activity with MTT and trypan blue dye.4. Establish the SD rat osteosarcoma model, and divide it into two groups, test group and control group. The tumor grows to 1.5X1.5cm a size roughly after two weeks, is even the iron magnetism nanometer flour to inject in to the tumor, after 0.5 hours, hand over to the 50 kHz magnetic field and irate 40 minutes. The tumor diameter was measured one week after irratation, and the animal wan put to death. The tumor was stripped intactly.
