The cartilage consists of the perichondrium and tissue, which including chondrocyte, matrix and fibre. It can be divided into hyaline cartilage, elastic cartilage and fibrocartilage by fibre contents in cartilage tissue. The hyaline cartilage is a typical cartilage with their special structure, so called it as a true cartilage on which cover articular and costochondral tissue. Fibrocartilage distributes to the intervertebral disc, articular disc, and pubis symphysis. Elastic cartilage distributes to auricle and epiglottis.The knee joint (KJ) cartilage is a typical hyaline cartilage, but temporomandibular joint (TMJ) cartilage is the fibrocartilage by thinking with what the category was synovial joint. A great quantity of research results have been showed, the fiber component or collagen types, proteoglycan in cartilage tissue of knee and temporomandibular joint are similar, moreover their functions and constructions are the same. But these research results are limited respectively in knee joint or temporomandibular joint study, little comparing research carried on. Hence, it is necessary to study on cartilage tissue property, matrix component and changing tendency of knee joint and the temporomandibular joint in an identical individual at the same time. In This thesis, at first ,we have examined morphological alterations in both knee and temporomandibular joint cartilage tissue section with hematoxylin-eosin(HE) staining, in hope of finds out their histology feature similarities and differences spots in two kinds of articularcartilages; Secondly, amino acid, aminohexose and water content of the articular cartilage have been analyzed, in order to explore divergence of upper mentioned components in KJ and TMJ, which are basis of histology. Finally, the study her was to characterize the presence and relative amount of decorin and biglycan in the condylar/tibial cartilage of the temporomandibular and knee joint by in situ hybridization ,and to assess their tissue localization by imrnunohistochemistry. Our aim was to supply experiment data for exploring mechanism of reconstruction and repairing of both articular cartilages. Meanwhile, to study age/sex-associated changes in decorin and biglycan in rabbit articular cartilage, we used 5-week-old and 7-month-old female and male rabbits in the entire experimental process.Part I :In the articular cartilage in TMJ and KJ of juvenile and adult rabbit, little procollagen in the interterritorial matrix, more extracellular, chondrocytes located in the cartilage cavities have been found and cartilage capsule surrounding cavities was distinct. The distribution of chondrocyte in the cartilage seems to be disciplinarian, The chondrocyte near the perichondrium is puerilism, small in diameter, flatted and located individually. In deeper zone including reserve, proliferating, calcified cartilage zone, cartilage capsule in which have a few cells and every zone is clear .All of these property demonstrate that cartilage in TMJ and KJ are hyaline cartilage.HE stain demonstrated that perichondrium on articular cartilage in the juvenile and adult rabbits TMJ were thicker than on KJ, disc of TMJ in every experimental group is distinctly visible , Express out the representative fibrocartilage histological feature. In the juvenile rabbit experimental groups, collagen fibre ranges irregularly, Orientation is not definite, diameter is homogeneous in disc; Every zone of cartilage is clear in condyle, zone of resting cartilage, proliferating cartilage, hypertrophic cartilage and calcified cartilage zone have been seen under microscope. In the adult rabbit experimental groups, every zone is clearer in condyle cartilage than in juvenile; chondrocytes in the disc succeeds line the range, that more chondrocytes are ranged against the fibre parallel in the cartilage capsule. These results show that 5-week-old rabbit is growing rapidly and its articular cartilage developing, and 7-month-old rabbit is developed.
