Objective To provide an easy ,quick and economical method to synthesize ofsiRNA , T7RNA polymerase is used,and the reactive condition is further optimized . To investigate the effect of RNA i. on the telomerase activity ,so to provide a possible way to cure the tumor .Methods Two RNA strands are synthesized separately using T7 RNA polymeraseand annealed to form siRNA. The siRNA is transfected into the tumor cells or added into the lysis of the tumor cells,and the interfering effect on the telomerase activity is judged by Silver-staining telomeric repeat amplification protocol(TRAP).Results T7 RNA polymerase can efficiently transcript the template with only partlydouble-strand .The RNA output is 8.6 n g in the 50 u L reaction system. siRNA can suppressed the telomerase activity notably, and much more efficiently than the same amount of antisense RNA strand.Conclusion The synthesis of siRNA with T7 RNA polymerase is a rapid ,efficient,high-output and economical way to synthesize siRNA. That RNAi can suppress the telomerase activity of tumor cell Maybe anew way to cure tumor.
