OBJECTIVES:In recent years, the GLN effect on intestinal mucosal barrier on the condition of trauma,infection and TPN were widely studied. Most of these studies showed GLN can protect the intestinal mucosal barrier. Radiation is a important therapeutic tool to some tumors. One of common complications induced by radiotherapy is the intestinal tract injury. On abroad though there were some reports on the GLN protective effect on intestinal mucosal barrier from radiation injury, the data about that were very rare and scattered. So the objective of our experiment is to deeply observe the protective effect of GLN on intestinal mucosal barrier of rats receiving radiation, and to make preliminary explore on the effect of GLN on protein metabolism , so as to confirm the GLN's beneficial effect on radiation injury animals and provide the experimental evidence for GLN clinical application.METHODS: 30 healthy male rats were randomly divided into three groups: control group,GLN-enriched group(+GLN)and GLN-free group (-GLN).The feeds of -GLN and control group were composed of rats' routine feeds plus 3% casein, while feeds of the GLN group were added into 3% GLN on the basis of routine feeds.After 14 days feeding, +GLN and -GLN group rats were received 9.0 Gry γ-irradiation in total body.On the twentieth day ,we killed all rats by cutting their iliac artery to obtain blood and Then we took the jejunum,MLNand serum to detect indexes below:1,jejunum mucosal morphometrics by light microscope and scanning electron microscopic (SEM).2,the rate of BT.3,IECs' DNA and protein content.4,serum endotoxin levels.5,serum GSH and SOD levels.6,serum TP,ALB,PA,RBP and TRF. Besides, we observed the BW changes in the whole experimental period.RESULTS:1, In -GLN group, intestinal mucosa was thin and mucsa atrophy occured,villus was short,In mucosa lamina propria plenty of lymphocytes were seen.SEM showed the top of villus had obvious ulcerations. In +GLN group the pathomorphologic injury degree was slighter than -GLN group. Quantificational analysis also showed the average villus height,average villus mass in +GLN group was much higher obviously than -GLN group.2, MLN bacterial culture results showed that intestinal BT rate of control group was 12.5%,+GLN group 22.2% and -GLN group 70.0%.the BT rates between +GLN and -GLN group had significant differences(P<0.01).3, The serum endotoxin level in control group was 249.7±23.44Eu/L,+GLN group 422.7±54.12 Eu/L,–GLN group 526.8±36.14 Eu/L.In three groups there existed significant differences.4, The serum GSH in control group,+GLN group and -GLN group were 11.18±2.74μmol/l,8.13±2.66μmol/l,4.684±1.38μmol/l respectively.The GSH levels of +GLN group and -GLN group were lower than that of control group ,whereas ,the level of +GLN group was obviously higher than that of -GLN group(P<0.01).The serum SOD levels in three groups were 177.11±5.68μmol/l ,176.4±6.65μmol/l,167.1±7.95μmol/l respectively. The level of +GLN group was higher than that of -GLN group (P<0.01).5, The IECs' DNA and protein contents in +GLN group had no significant differences with control and -GLN group.While between -GLN and control group there was difference on the IECs' protein content(P<0.01).6, The BW in three groups all had increased positively before radiation.Comparing the BW on the 1st day before radiation with on the 5th day after radiation,we found the BW changed considerably: +14.0±4.14g in control group , -28.5±6.59g in +GLN group and -36.5±8.83g in -GLN group.It shows after radiation the BW in +GLN and -GLN groups were obviously lower than control group,but the BW in +GLN group was much less than -GLN group.The serum TP level had no difference between +GLN and -GLN group.The serum ALB in -GLN group was lower than control group(P<0.01),while no difference was seen between +GLN group and control group.Similarly,the serum RBP and PA levels in -GLN group were both lower than control group(P<0.05,P<0.01).
