Objective: To detect the expression of MUC1(mucin1)and TGF-β1 (Transforming Growth Factor-beta 1) in the endometrium during menstrual cycles and investigate the relationship between the expression of MUC1/ TGF-β1 and the serum levels of E2,P during peri-implantation period.Methods: The biopsy specimens in 60 samples of reglular menstrul cycle and ovulation normality women's endometrium were processed to determine the expression patterns of MUC1/ TGF-β1 by immunohistochemistry and pathologic image semi-quantification techniques. The pinopodes, which were used to be an indicator to signal "implantation window", can be observed in the endometrium of mid-secretive phase by SEM(Scan electronic microscopy). Serum E2 and P during peri-implantation period were measured to investigate the relationship between E2/P and MUC1/TGF-β1 expression to evaluate the impact of E2/P on MUC1/TGF-β1 expression during peri-implantation period.Results: 1. MUC1 expression was exclusive to the epithelium cells(luminal and glandular epithelium) of the uterus under all conditions examined.MUC1 expression was lower in preliferative phase, but obvious higher in secretive phase. It was significantly higher in mid-secretive phase,especially during peri-implantationperiod(P﹤0.05),decreased in the late-secretive phase. 2.TGF-β1 can be observed in luminal,glandular epithelium and stromal cells in the different phases of menstrual cycles. TGF-β1 expression was lower in proliferative phase, but obvious higher in secretive phase, especially higher during peri-implantation period(P﹤0.05). 3.There were statistically positive relations between the serum P levels and MUC1 expression in the endometrium,also positive relations between the serum P levels and TGF-β1 expression in the endometrium(r=0.6,r=0.555,P﹤0.05,respectively) during peri-implantation period. 4.There were no significantly correlations between the serum levels of E2 and the expressions of MUC1 orTGF-β1 in the endometrium(r=0.367,P>0.05;r=0.197,P>0.05)during the same period.Conclusions: The high expression of MUC1 in the ovulation normality women's endometrium during peri-implantation period may be closely related to the receptivity of the endometrium; 2. The high expression of TGF-β1 in the ovulation normality women's endometrium during the secretive phase, especially high expression during peri-implantation period, may be also closely associated with the receptivity of the endometrium;3. MUC1 and TGF-β1 expression seems to be regulated by the ovarian steroid hormones E2 and P.
