| Atherosclerosis(AS) is main pathological foundation of cardio-cerebral angiopathy that is a kind of disease threatening to human health most.Domestic and internat- ional medicine has not been satisfied with treating it yet. From the pathogenesis generally acknowledged,there are several kinds of therapy, such as regulating serum lipid, anti-oxidant, prenenting vessel endothelium from bing damaged, resisting thrombus not to form, etc. Those ther- apies have a great progress than ever before, but some side effects, high recurrence rate and death rate still remain high. To study and find a kind of effectual remedy is very imperative.According to the disease's characterstic and clinical manifestation, AS should be the range of Traditional Chinese medicine (TCM) [1-4] "heart pain" "chest pain" "palpitation" "vertigo", etc. The pathogenesis should be Tan, Yu, Xu. There are a large number of effective Chinese herbal medicine and prescriptions recorded, systematic and profound understanding about it in TCM. How to explore and develop those Chinese prescriptions to prevent and cure AS is already rather important.DangguiShaoyaoSan(DSS, from《Jinguiyaolue》), consisting of Danggui Shaoyao Chuanxiong Fuling Baizhu and zexie,has the effects of "Regulate the blood and remove blood stasis ,Clear sputum and resolve muddy, Invigorate the spleen for excreting the dampness". Following the principle of "Strictly adhere to the pathogenesis and treat the different disease in the same way", we selected DSS to prevent and treat AS by animal experiment study under the guidance of clinical experience of Professor YanYanli, trying to offer for a kind of ideal Chinese medicine treatment and the reliable experiment for clinical practice.Objectives: To observe the effects of DSS on experi- mental AS rabbits, and to study the mechanisms of DSS preventing and treating AS.Methods: According to the orginal prescription's quality proportion (Dangugi:Shaoyao: Chuanxiong: Fuling: Bbaizhu:Zexie is 3:16:8:4:4:8), If we take the clinical usual quality as 10g, the other fives should be calculated in the former proportion , and then according to "the proportion of human-rabbit body surface area"[5], The medicine quality of low-dosage group: crude drug 24g/kg.d, high-dosage group: crude drug 48g/kg.d.According to the relevant document[5], the model of experimental AS rabbits was established by feeding the cholesterol contained fodder for 8 weeks. Fifty healthymale New Zealand rabbits weighing 1.5kg-2.5kg were devided into five groups, and each consist of ten. The five groups are as follow as:(1) Control group , the rabbits are fed with the normal fodder. (2) Model group ,the rabbits are fed with high-fat fodder, which contain 1% cholesterol, 4%yolks powder and 10% lard in the normal fodder. (3) DSS-Low dosage group, the rabbits are fed with high-fat fodder added DSS, there is 48ml concentrating liquid of DSS in each kg's high-fat fodder,which equals to 480g crude drug. The DSS liquid should be mingled with the high-fat fodder even in the course of the processing. (4) DSS-high dosage group, the medicine quality is two twice than that in the DSS-low dosage group. (5)Positive medicine contrast group(Jifeiluoqi), the rabbits are fed with the high-fat fodder added Jifeiluoqi, 0.15%Jifeiluoqi contained in the high-fat fodder. Every rabbits in each group is fed with equal amount of fodder, namely100g.Separately on 2th week, 4th week,8th week,serum TC TG, HDL are measured and LDL ,AI are calculated by the fowlling formula: LDL-c=TC—(1/5TG+HDL-c), AI=(TC—HDL-c)/HDL-c. After collecting blood, the rabbits were sacrificed by embolism, separate and take down all aorta, four sample were selected randomly from each group, which is 2cm part of originated aorta, and put them into 70% alcohol after being washed away the clot in PBS.
