U.urealyticum which lacks of cell wall, belongs to the family mycoplasmataceae and differ from other mycoplasma species by its ability to hydrolyse urea, meanwhile it is the smallest, self-replicating prokaryotes The prevalence of colonization of urinary tract of normal men has been reported as approximately 25%, Rates in women vary from 0 to 80%. There are relatively few reports on U urealyticum pathogenesis. In man, U urealyticum has been showed to cause approximately 15 %of no-specific urethritis (NSU). It can cause decrease in lateral head displacement and sperm mortality, and the latter may result in reduced egg penetrability In pregnant women, U.urealyticumis a significant cause of chorioamnionitis, frequently with a sequela of adverse pregnancy out-come. U urealyticum may be acquired by neonates either in utero or by vertical transminition at birth, and can cause pnemonia, pulmonary hypertension, chronic lung disease and meningitis of the new born. Recently with the further insight of STD, U.urealyticumhas been noted in association with sexually transmitted diseases.A 14-member serotyping scheme has been widely accepted. On the basis of several criteria, the serovars can be divided into two distinct clusters or biovars. Because of the serotyping of U.urealyticum has no help to determine its clinical pathogenicities and can not determine the relation of some serotypes to some special clinical process, recent moleculer characterization of the U.urealyticum serovars has led to an effort to reclassify them into two species, U.urealyticum would include 10 large genome serovars(0.88~1.2Mbp). U.parvum would includethe small genome servers 1, 3 , 6, 14,(0.75~0.76Mbp). The 5-end region of MBa gene can subtyping the biovars. Through which U.parvum was separated into three subtype : subtype 1 includes serovar 1, subtype2 includes serovar 3/14, subtype 3 includes serovar 6. Meanwhile U.urealyticum was separated into three subtypes too.Till then, U.urealyticum was genotyped by PCR-RFLP and the relation between genotype and disease are discussed. However because of its complication and expensive,even the difficulty of defmitethe result, it's refrain used widely. So its important to develop some simple and economical method to genotype the clinical specimen.In our research, we developed a compound-PCR method, through numerous experiment, we improve the annealing temperature at 58 and the sequence DNA concentration at 8 l,and then using this method to investigate the moleculer epidimiology of U.U . The result show that all the specimen can be separated into two species,and U.parvum 60 (60/61 98.36%), U.urealyticum 1 (1/61 1.64%).And then genotyping the U parvum, in 60 specimen, typel is 3 (3/60 5.00%), type 3/14 is 57 (52/60 86.67%), type 6 is 1(1/60 1.67%), and 4 (4/60 6.67%) specimen are compound types.Of the 61 patients,16 cases followed with clinic symptom and sign (26.67%), 3/14 and Itype show the highest rate.The compound-PCR method of our research is more convenient, economical and easy to use, it is more practical than other methods used domestic, and easy to use in clinic diagnose and research.
