PrefaceTransforming growth factor - pfamily is a kind of isodimer poly-peptide belonging to structural associated protein. Up to now, three kinds of homology isomers, TGF -β1, TGF -β2, TGF -β3 , have been isolated from mammalia cells and they all have similar biological activity. TGF -β1 is the main form in human body and it can affect the development, differentiation, adhesion and immigration of cells, the formation of vessel and extracellular matrix and immunity function. It has been found that TGF - β1 with high activity and expression has association with the metastasis of lymph node. E - cadherin is a kind of calcium dependent adhesion molecule. It locates in the normal cells and mediate the homotype adhesion between cells, so it plays an important role in the formation and maintenance of tissue structure. It has been shown that the decrease or loss of expression of E - cd in many malignant tumors is associated with the infiltration and metastasis of cancer cells. Based on the theory, we applied SABC immunohis-tochemistry on 60 cases of paraffin samples of SGLSCC from 1994 to 1996 to detect the expression of TGF -β1 and E - cd in SGLSCC and analyzed their biological relation in order to evaluate their diagnosis value in the metastasis of cervical lymph nodes and their prognosis.Experimental materials60 paraffin samples were collected form the patients with SGLSCC who hospitalized in the ENT department of the first affiliated hospital of China Medical University from 1994 to 1996. 10 normal samples of the same time were also been collected. There were 37 cases of male and 23 cases of female, aged from 36 to 79, the mean age was 58. 1 years old. All the patients were followed up more than 5 years, in which 9 cases died in one year after operation, 20 cases died in one to five years after operation and 25 cases lived up more than five years. According to the criterion of UICC, the clinical staging was 3 cases of stage I , 13 case of stage II, 21 cases of stage III, 23 cases of stage IV; the pathological grading was 18 cases of stage I (high differentiation ) , 18 cases of stage II ( medium differentiation) , 24 cases of stage III( low differentiation) . In all the samples, 34 cases had lymph node metastasis and 26 cases didnt have. Two groups were divided according to the patients'sex, age and the size of tumor.Experimental methodsAgents and methods: TGF-β1 poly antibody (V) were purchased form American Santa Cruze corporation; E-cd poly antibody, SABC Test Kit and DAB kit were purchased from Boster corporation. All the reagents were ready to use. SABC immunohistochemistry were applied in this study with PBS as a negative control and positive slide supplied by corporation as positive control.Statistics methods; Chi -squared test, Kendalls ranked correla-tion analysis and Kaplan - Meier analysis were used and processed by SPSS10.0 software.Experimental results1. The positive expression rates of TGF -β1 and E - cd in 60 cases of supraglottic laryngeal squamous carcinoma were respectively 81. 7% (49/60) and 70% (42/60) , while in normal laryngeal mucosa they were only 20.0% (2/10) and 90% (9/10).2. The relationships of the expression of TGF - β1, and E - cd in pathological grading and clinical stage were respectively positive correlation and negative correlation. These ranked correlation had statistic significance ( p <0.0001).3. Both expression rates also had significant statistic difference in the group with lymph node metastasis and the group without (p <0. 0001) , while in the groups with different sex, age and tumor size, the difference was not significant ( p > 0.05 ) .4. After statistical analyzing, the two indexes had no concordance (Kappa = -0. 198, p=0. 077) , that meant there was no coexpres-sion relationship.
