Objective To investigate the expression and relation of extracellular signal-regulated kinase-l(ERKl) and cyclin Dl in acute leukemia(AL).Method The expression of ERK1 and cyclin Dl were detected in bone marrow cells from 44 cases of newly diagnosed or relapsing acute leukemias(AL) patients and 13 cases of complete remission AL (AL-CR) patients and 15 cases of controls by immunohistochemistry stain . Positive cells were dyed yellow or brown in nuclei. Positive ERK1 or cyclin Dl was that the positive cells were >5% bone marrow cells.Results 70.4% AL patients were positive for ERK1 and 59.1% AL patients were positive for cyclin Dl, both were remarkably significant higher than AL-CR group and control group(P<0.01). The expression of ERK1 and cyclin Dl have no difference between acute lymphoblastic leukemia(ALL) and acute non-lymphocyticleukemia(ANLL) (P>0.05). The expression of ERK1 have no difference between high-risk group and low-risk group in childhood (P>0.05), but the expression of cyclin D1 in childhood high-risk group is significantly higher than that in low-risk group (P<0.05) . Significant positive correlation between the expression of ERK1 and cyclin Dl was found in AL (r =0.677, P<0.01).Conclusions (1)The rate of overexpression of ERK1 and cyclin Dl was quite high in AL patients.(2)Cyclin Dl may be related to prognosis in childhood patients of AL. (3)ERK1 and cyclin Dl could play important role in the development of AL.
