| ã€Objective】 To investigate the expression of three angiogenic factors VEGF121,bFGF,PD-ECGF in gastric carcinoma and whether tumour microvessel density (MVD) immunoreactivity and clinicopathological features of gastric carcinoma are related to the expression of VEGF121,bFGF,PD-ECGF.ã€Methods】 Immunohistochemistry technique (SP and SABC) was used to test the expression of three angiogenic factors and CD34 in 59 gastric carcinoma specimens ,12 normal gastric membrane beside carcinoma specimens and 25 gastric benign lesions specimens embedded in paraffin wax . VEGF121,PD-ECGF, CD34(assayed by SP) were of mouse monoclonal antibodys and bFGF was of rabbit polyclonal antibody (assayed by SABC ), the CD34 antigen was carried out to evaluate MVD in tissues. Negative controls were included by replacing the primary antibody with the PBS. Statistical analysis was performed using the PEMS software package, with p < 0.05 being considered significant.ã€Results】 â‘ Immunoreactivity for VEGF121 was found in cytoplastic and cell surface of most gastric carcinoma cell (positive staining 47 in 59),the rate of positive staining in gastric carcinoma was the highest(p<0.01); There were 39 in 59 which were seen PD-ECGF positive staining, the rate of positive staining in gastric carcinoma was also the most highest(p<0.01); bFGF positive staining was detected in 39 case(39 in 59), the positive rate in gastric carcinoma group was the hightest among three groups,but the difference was insignificant(p>0.05); the MVD of gastric carcinoma was higher than that of normal gastric tissue beside carcinoma and that of gastric benign lesions(P<0.01),Moreover,the shape and structure of microvessel was abnormal.â‘¡ MVD of VEGF121 or bFGF positive staining group was higher than that of respective negative staining group(p<0.05),and the expression of VEGF121 or bFGF was positive correlation with MVD(p<0.05). â‘¢Farthermore infiltration depth and distance metastasis were positive relation to the expression of VEGF121(p<0.05),inspite of p>0.05,the rate of positive staining in the group with lymph node metastais was higher that in the groupwithout lymph node metastais; Yet we didn't discover bFGF was in association with clinicopathological features of gastric carcinoma . PD-ECGF wasn't in association with infiltration ,lymph node metastasis and distance metastasis of gastric carcinoma too,but man case and adenocarcinoma case have higher PD-ECGF positive staining(P<0.05). â‘£55 in 59(93.05%) at least express one of three angiogenic factors; VEGF121 was positive correlation with bFGF(P<0.05),but we only found there was a tendency that the speciments with both VEGF121 and bFGF positive staining had higher MVD than the speciments with one positive staining,the difference wasn insignificant(P>0.05);Both VEGF121 and bFGF weren't correlation with PD-ECGF(P>0.05); Similarly we only found there was a tendency that the speciments with three factors positive staining had higher MVD than the speciments with two positive staining,but the difference wasn in- significant(P>0.05).ã€Conclusions】①The expression of VEGF121and PD-ECGF is higher ,moreover the number of microvessel in gastric carcinoma is larger and the shape and structure of it are abnormal.â‘¡Both VEGF121 and bFGF can improve vascularization in gastric carcinoma,it is possible that vascularization in gastric carcinoma is blocked up by means of restraining expression of VEGF121 and bFGF.â‘¢ VEGF121 is the most important for vascularization in gastric carcinoma among three factors ,the next,bFGF is useful too. Significance of PD-ECGF expression in gastric carcinoma isn't clear,so obstrucion of vascularization in gastric carcinoma had better aim at VEGF121.â‘£ VEGF121 is positive correlation with infiltration and metastasis of gastric carcinoma,measurement of VEGF121 expression may be value to evaluate clinical progression of gastric carcinoma. |
