Objective Nabumetone(NAB),a selective cyclooxygenase-2(COX-2) inhibitor ,belongs to non-steroidal anti-inflammatory drugs (NSAIDs) and was used to treat osteoarthritis(OA). Total glucosides of paeonia(TGP) is the effective composition extracted from the root of paeonia Lactiflora.lt possesses the effects of anti-inflammation,analgesia and immuno-regulation.According to the difference of the pharmacological character between two drugs,the therapeutic effects and safety of NAB alone or combined with TGP will be investigated in this study. Methods The 57 patients with different stage knee osteoarthritis(OA) were divided into two groups randomly and treated by NAB and NAB+TGP respectively.In NAB group,27 patients (male 7,female 20,age 57.04 ?8.14 y) were given NAB 1000mg(po,qd) for 8wk.The other 30 patients (male 3,female 27,age 55.33 ?. 91y) were given NAB 1000mg(po,qd) and TGP 600 mg(po,bid) for 8wk.They were observed regularly.All these data were figured out and analyzed by Epi info.Results The total efficacy rates of NAB and NAB+TGP were 22.22% and 26.67% respectively for four weeks treatment,and 48.15% and 70% respectively for eight weeks treatment. Both in the 4th week and the 8th week,there were significant differences in pain on pssive m-otion,15-m walk time and the patients assessment between two groups knee osteoarthritis. Also in the 8th week,there were significant differences in joint tenderness and between two groups. There is no significant differences except in the case of stomachache among adverse reaction rates of the two groups.Conclusion NAB combined with TGP is more effective safe than NAB in treatment of patients with knee osteoarthritis.
