| Objective: Studying the effect of Jian-Gu-Er-Xian Extraction on rats' osteoporosis. Methomd: Three experiments were done to study the topic. O 16 6.5-month-old female Wistar rats were randomly divided into 2 groups: Sham operation group (Sham) and ovariotomy group (OVX) . Sham group were sham operated while OVX group were ovariotomized. 6 weeks after operations, weight, bone density (BMD), blood calcium, blood phosphorus (P), blood alkalescence phosphate enzyme (ALP) and blood tartrateresistant acid phosphatase (TRAP) were detected. ?42 6.5-month-old female WA/Jic Wistar rats of SPF/clean grade, inbred strain, were randomly divided into 4 groups: Sham operation group (Sham) with 11 rats, ovariotomy group (OVX) with 10 rats, ovariotomy group given Premarin (Premarin) and ovariotomy group given Chinese herb, Jian-Gu-Er-Xian extraction (Herb). The Sham group was sham operated while the others were ovariotomized. 6 weeks later, sham group and OVX group were given 0.9% NaCl 1.6ml ?kg"Jper rat. Premarin group were given premarin 0.065 Img ?kg"1 per rat. Herb group were given Jian-Gu-Er-Xian extraction 1.85ml ?kg"1 per rat. All the animals were sacrificed after having been treated for 10 weeks. All the above parameters would be detected and biomechanical test for left thighbone and the fifth lumbar would be done. ?14 18-month-old male SD rats were randomly divided into 2 groups: blank comparison group (BL) with 7 rats, group treated by Jian-Gu-Er-Xian extraction (Herb) with 7 rats. BL group were given 0.9% NaCl 1.6ml ?kg^per rat and herb group were given Jian-Gu-Er-Xian extraction 1.85ml ?kg"1 per rat. After treating for 10 weeks, left thighbones were taken to do biomechanical tests. Result: O 6 weeks after operations, compared with sham group, weight, Ca, P, ALP, TRAP, BGP of OVX group had increased while BMD had decreased. ?Compared with sham group, weight, Ca, P, ALP, TRAP, BGP of OVX group had increased, while the biomechamics capability and BMD had decreased. Compared with OVX group, the weight of Premarin group and herb group was partly controlled, while ALP, TRAP and BGP decreased and biomechamics capability improved. f> Compared with BL group* the data of the max shearing stress of herb group was evidently stronger than that of BL group. Conclusion : The symptom of postmenopausal osteoporosis (PMOP) can be successfully simulated 6 weeks after ovariotomy. Jian-Gu-Er-Xian extraction can treat postmenopausal osteoporosis with the close effect of Premarin. Jian-Gu-Er-Xian extraction can improve the biomechanical capability of cancellous bone of aged rats to some degree, so it may prevent senile osteoporosis. |
