| Objective To investigate the expression of midkine(MK) andmicrovessel density(MVD) in breast carcinoma in relation to biological behavior and prognosis .Method Expression of midkine protein and microvessel density levels were detected by immunohistochemical technique in 67 breast invasive ductal carcinoma specimens. The relationship of MK and MVD expression to clinicopathological characteristics and postoperative survival time was statistically analysed .Results (1) Of the 67 cases , MK immunoreactivity was positive in37 (55.2%) . MK expression level was associated with tumor size , axillary lymph node status , local recurrence and distant metastasis . Univariate survival analysis showed that the postoperative 5-year survival of patients with positive MK expression was significantly higher than those with negative MK expression (P<0.05) . (2) All 67 cases were divided into high MVD group and low MVD group . The number of MVD was correlated closely with tumor size , axillary lymph node status , local recurrence and distant metastasis . Univariate survival analysis demonstrated that patients with high MVD showed worse postoperative 5-year survival (p<0.01) . (3) A significantly higher MVD (90.75+33.05) was observed in the tumors with positive MK expression compared with tumors with negative MK expression (70.48 + 31.33) (p<0.05) . Conclusions (1) MK expression is closely correlated with tumor development progression and prognosis in breast carcinoma, so may serve as a valuable tumor marker and prognostic indicator. (2) IncreasedMVD in breast carcinoma promotes malignant progression, and associates a poor outcome. MVD is an important prognostic factor in breast carcinoma .
