OBJECTIVE: To investigate the effects of Chinese herbal medicine(CHM), LCF-2 prescription, and modem medicine(MM) used alone and combination in the physiology, immunology, morphology on the BXSB mice and the mechanisms of treatment to SLE.METHODS: BXSB mice were randomly divided into 4 groups: group A was model; group B was CHM; group C was MM; group D was CHM+MM. Weight and mortality of the BXSB mice were dynamic observed. Spleen index and thymus index were measured by a method of weight. The levels of proteinuria in the different groups were measured by Four Bromophenol Blue. Serum dsDNA^ IL-2, IL-6^ TNF-a were measured by ELISA. IgG and complement C^ in renal glomeruli in all BXSB mice were analysed by directly or indirectly immunofluorescence. The changes of morphology were also observed.RESULTS:1. In Groups B and D, the weights of BXSB mice were remarkably increased in comparison with model group (p<0.05). Mortality in group B was 0, which was significantly different from group A (p<0.05). Although mortality in group D was also lower than that of group A, there was no significant difference in statistics. The levels of proteinuria in group B, C and D were all lower than that of group A (p<0.05).2. 2. The thymus index in Group B was higher than that of controls, but there was no difference in spleen index. Group C could decrease both of spleen index and thymus index, but group D could reduce spleen index- 4 -(p<0.01) and increase thymus index (p<0.05). The level of anti-dsDNA antibodies decreased remarkably in the groups of modern medicine, which could convert it into negative. The percentages of CDa+Tx CD4+T^ CDg+T and CD4+T/CDg+T raised remarkably in group B and group D. However, group C could significantly down-regulate the percentage of CD4+T cells, but had no effects on CD3+T and CD8+T. Therefore ratio of CD4+T/CDg+T reduced, which indicates the mechanism be different between group B and C. As to the cytokine detection, CHM could significantly down-regulate the concentration of TNF-a (p<0.05) and up-regulate the concentration of interleukin-2 (p<0.01) albeit it has no effects on interleukin-6. In contrary, modern medicine in group C could significantly inhibit production of interleukin-6 and TNF-a, and no effect on interleukin-2.3. The fluorescence intensity of IgG and €3 in the kidney of BXSB mice were weaken in group B, C (PO.05) and D (PO.01) compared with group A. The group D obviously alleviate the renal pathologic damage in BXSB mice.CONCLUSION: Our results indicate that CHM and MM. should be effective on BXSB mice albeit the mechanism was different. The CHM and MM used in combination was superior to that of alone. |
