Experimemtal Study On Arsenic Trioxide And All-trans Retinoic Acid's Effects On Neuroblastoma Cell Line SK-N-SH And Nude Mice Transplants Tumor

Objective: ㏕o study the role of Arsenic trioxide(As2O3) and All-trans retinoic(ATRA) inhibiting cell proliferation and inducing apoptosis in Neuroblastoma(NB) SK-N-SH cell line in vivo and in vitro;(2)To set up NB transplanting model in nude mice and study it's biological characteristics. Methods:(1)ln vitro the effect of AsiOs and ATRA inhibiting cell proliferation was assessed by Mtt assays, and the apoptosis index (AI) was detected by Tunel-Pod method ;(2)Using cell transplantation and tissue graft to set up NB transplanting model in nude mice.Three generations of the model was recorded; (3)To observe the tumor growth rate > volume and weight in the As-iO^ or ATRA treatment group and in control group.(4)In vivo,the effect of As2C>3 inhibiting cell proliferation was assessed by detecting proliferating cell nuclear antigen (PCNA)-positive nuclei cell index (PI) , and the apoptosis index (AI) was detected by Tunel-Pod method.Result: (1) In vitro,all concentration groups can inhibit cell proliferation,extending act time can improve the effect more than that of increasing drug dose.,and all concentration groups can induce apoptosis ,AI was higher in the high concentration group.(2)In vivo,the tumor growth rate was decreased in the As2O3 or ATRA treatment group, the rumor volume inhibition rate was 35.99%. 44.30% and weight inhibition rate was39.85%% 46.04%.(3) Comparing the control group,AI was higher (P<0.05) and PI was lower (P<0.01) in the As2O3 or ATRA treatment group.(4)Compare with cell transplantation,-3-using tissue graft to set up NB transplanting model in nude mice has more effective. The tumor growth rate has more shorter (P<0.01);The generate ratio of transplanted rumor has more higher(P<0.01).Conclution:(1)As2C>3 and ATRA can inhibit cell proliferation and induce apoptosis in Neuroblastoma SK-N-SH cell line in vivo and in vitro. They may be new promising drugs to treat Neuroblastoma. ㏕he nude mice NB's tumormodel using tissue graft is suitable for NB's study.
