Objective To observe the effects of selenium (Na2SeO3) on the apoptosis and proliferation of cuLtured k562 cells, the activity of erythrocyte adhere to S180 cells and natural killer cell (NK cell) and to study the mechanism of selenium on the apoptosis.Methods Using the technique of cell cuLture and the medium being exposed to different doses of Na2SeO3 (0~10.00μmol/L) , the apoptosis and proliferation of k562 cells were examined by means of Fluorimetry and MTT, while the adherence activity of e-rythrocyte to S180 cells and the killing activity of natural killer cell (NK cell) were determined using immune adherence assay and modified MTT.ResuLts Different concentration of Na2SeO3 (0 ?10. 00/Jmol/L) can increase the apoptosis rate of K562 cells with a relationship of dose-effect response. The concentra-tion(0. 10 ?0. 50/imol/Dof Na2SeO3 can inhibit the proliferation of k562 cells, enhance the adherence activity of erythrocyte to S180 cells and the killing activity of NK cell, with these activities reaching to the peak in the concentration of 0. 50/imol/L. While the concentration (5. 00 ?10. OOpimol/L) can erythrocyte adherence activity and NK cell killing activity, with these activities being significantly lower than those of the control groups by the concentration of 5. OOpimol/L.Conclusion Selenium can induce apoptosis of cuLtured k562 cells with a relationship of dose-effect response, it may play a important role in the process of apoptosis of tumor cells by means of inducing cell apoptosis, inhibiting cell proliferation and enhancing erythrocyte adherence activity and NK cell killing activity to tumor cells.
