Purpose: To examine and evaluate the ability of spontanousregeneration following various extent of incomplete injury in aduitrats.Metbods: The quantitive and reduplicated varius extent incompleteinjuried model of adult rats optic nerve was established using differentwounding time with constant wounding force produced by acrossaction forceps at 2mm behind eyeball. GAP-43 (growth associatedprotein-43) and its mRNA erpressions wer detected withimmunhistochemistry (IHC) and reverse transcription polymerasechain reaction (RT-PCR); The site and processes of spontaneousregeneration wer observed by means of anterograde tracing offast-DiI, high affinity fluorescent dye to the neuron and axon, Whichinjected into the vitreous cavity, and the changes of ultrastructure wasviewed by eletron microscope at different recovery time pointsfollowing various extent incomplete injury.
