Xuanhusuo Powder is composed of Yanhusuo and Glycyrrhizae Radix of equal weight, used to cure cardiodynia. In this paper, a systematic study on the compound chemical composition and pharmaceutical effect was done by modern technique in order to illuminate the principle of Xuanhusuo Powder and provide scientific basis for clinic application. The total Alkaloids of yanhusuo and Glycyrrhizic acid is the effective part of Xuanhusuo powder in myocardial injury induced by Iso in rats and mice. Preparation methods have different effect on the model of myocardial injury rats. Jon-pair extraction combined with UV-spectrophotometry and HPLC have been respectively applied to determine the content of several Xuanhusuo Powder prepared by different methods. Preparation methods have different effect on the content of the total alkaloids, tetrahydropalmatine and glycyrrhizic acid. The precipitate formed during decocting of the Chinese herbal complex prescription Xuanhusuo powder is studied in chemical and pharma- cological part. Yanhusuo alkaloids and glycyrrhizic acid were precipitated off the decoction. The precipitate possessed any pharmacological activity. It can inhibit the experimental myocardial ischemia and rabbit platelet aggregation induced by adenosine diphosphate, arachidonic acid with a dose-dependent manner in vitro. The mechanism of these effects might be related to calcium iron antagonism. To make clear the chemical mechanism of the precipitate, we manufacture the Tetrahydropalmatine-Glycyrrhizic complex salt. The salt, useful as a remedy for myocardial ischemia without bitter taste, by the treatment of tetrahydropalmatine with glycyrrhizinic acid was described. The complex salt, white powder, m.p.208 10 C. Tetrahydropalmatine-Glycyrrhizic complex salt could reduce the lipid peroxidation in the doxorubicin toxic cultured cardiac cells. |
