A lot of clinical observations and experimental studies have shown that prescription for invigorate the kidney and activate the blood may be used to lead to new bone formation and to enhance fracture healing. However , it had few reports and not been recognize well about the therapeutic mechanism promoted fracture healing with traditional chinese herbs. Further research should be performed such as how to regulate the growth and proliferation of osteoblasts in vitro, how to stimulate bone cytokine in vitro and intro with invigorate the kidney and activate the blood? In order to investigate the their mechanism of promoting bone healing. Experiments of three pats were carrid as follows: Part I :Effect of Kindney Invigoration and Activate the Blood on IGF- I in Experimental Fracture Healing Process Objective To observe the effect and influence of kidney invigoration and activate the blood on insulin like growth factor- I (IGF- I ) in fracture healing process in the rat. Method: Twenty four 7-month-old SD rats were randomized into two groups of 12 each: the therapy group and the control group. After the experimental model of fracture by operation was established,the therapy group was treated with Guyu tablet (GYT) 20g/kg body weigh/day for 28 days continuously by oral,whereas the control group was treated with equivalent sauna. Serum IFG- I concentration was dectected and histologe, radiologe and bone mineral density (BMD) as well as biomechanical strength of callus were measured at 2,4 weeks. Results : Serum IGF- I concentration of the therapy group was highter than that of the control group (PK0. 05, <0. 01) at 2th and 4th weeks. At 4th week,BMD in callus area and biomechanical testing of the therapy group were higher than that of control group. Conclusion: The function of kidney invogoration and activite the blood migh accelerate bone healing and it is one of machanism & promoting fracture healing withthat might parthy increase IGF- I in rat with fracture. ?? Part I :Effects of Guyupian on the Proliferation of Fetal Rat Calvarial Osteoblastic Cell in Vitro Objective: To investigate the effects of kidney invigoration and promote blood circulation on osteoblastic cell metabolism. Methods: Fetal rat calvarial osteoblastic cell were cultrued in the medium containing different concentrations of Guyupian (GYP) and incubated for 3 days. Calorimetric MMT ,3H-thymidine incorporation were detected to determine the condition of cell proliferation and differentiation in vitro. Results GYP promoted the proliferation of osteoblastic cells. Conclusion: GYP might promote the bone formation.
