| Cryptosporidiosis is a zoonotic protozoan disease of worldwide distribution, affecting a wide range of vertebrate hosts. Cryptosporidium from kids mainly causes diarrhea, even death. The epidemiological survey of calf Cryptosporidiosis were carried out in Henan Province. nested PCR-RFLP . So in order to understand the epidemiology and the species or genotype of Cryptosporidium from calves in Henan Province,In the present study, the epidemiology investigation of Cryptosporidium in calves were conducted in Henan province, then the Cryptosporidium isolates obtained were identified according to the morphology of oocysts, transmission study, genotypic analysis of 18S rRNA, nested PCR-RFLP.In order to understand the infection of intestinal parasites in cows, especially zoonoses parasitosis. we examined 154 feceal samples from two cows farms in Shangqiu using Lugol's iodine staining and Sheather's sugar flotation technique and acid-fast-method. The results showed that 116 samples were positive of intestinal parasites and the average rate was 74.68% (115/154). In which,the rate of Giardia infection was 7.14%(11/154); the rate of Cryptosporidium infection was 10.39%(16/154);the rate of Eimeri infection was 66.23%(102/154);the rate of Strongylus was 3.24%(5/154);the rate of Trichuris was 5.19%(8/154). The survey for clinical treatment and prevention of intestinal parasitic diseases dairy provides a reference and a basis.To establish a calf model of Cryptosporidium parvum. Neonatal calves were challenged orally with oocysts C.parvum which collected in this survey. Results showed that the patent period was in 2 days post infection,and the counted number of oocysts increased obviously, its excreted peak numbers of oocysts at the 5th days post infection,and persisted for 6 days.After dissected,the whole digestive tract mocosa were stained,oocysts were detected only in small intestine.Neonatal calves were successfully infected with C. parvum and established the Neonatal calves model for C. parvum.To understand the prevalence of Cryptosporidium infection in cows in HeNan and to assess the potential role of cows as a source for human cryptosporidiosis,2 species Cryptosporidium from calf feces was detected by analysis of the 18SrRNA gene was used.Then, Both Blast and Fasta methods was used to search homological sequences in NCBI, DDBJ and EMBL, after that, homological sequences were alignmented. PCR products were digested by SspI and VspI were identital to RFLP restriction maps of C.parvum of calves and different with C. baileyi obviously. Phylogenetic analysis showed that the Cryptosporidium species formed two clades. 0ne clade contained C. serpentis, C. andersoni and C.muris.The other clade contained C. bailyei, C.bovis,C.ryanae, C.meleagridis, C.hominis, C.parvum and two isolates from calves. In the second group, the two isolates formed a single taxon with the C. parvum. The present study revealed that calves commonly infect many zoontic parasites; calves can infect zoonotic Cryptosporidium species/genotypes and subtypes, thus, calvess may play a role in the zoonotic transmission of Cryptosporidium. |
