| Ggrape root is the the worst cold resistance organ of dormant grape,the root cold tolerance is a key factor for grape surviving in cold winter, Therefore, studying the cold hardiness of grape root has great significance.In this experiment, four year old cuttings of grape root varieties and grafted root for the test materials.Seted five temperatures, deal grape root with cold acclimation and de-training. The experiment contents include cell membrane permeability, structure observation and cold-related substances determination. Through these methods investigate genotype, cold acclimation, and grafting on the cold hardiness of grape roots.The results:1.Root cold tolerance of four grape varieties from strong to weak order is Beta>Jing Ya>Red Earth>Centennial Seedless, BeTa root was stronger than the other three species,Jing Ya root cold tolerance was slightly stronger than Centennial Seedless and Red Earth root.There was no significant difference between the latter two.2. Beta root as cold tolerant cultivar has Prominent Cold characteris.They include: Cold tolerant organizational structure, lesser membrane injuryed when under low temperature stress,lower water content, higher content of oleic acid percentage in biofilm.In low temperature stress, the content changes of proline, malondialdehyde, soluble sugar and soluble protein are more helpful to against low temperature damage.3.Cold acclimation can increase the cold hardiness of grape roots. They include: Enhance the stability of cell membrane, the increasing content of proline, soluble sugar and soluble protein is helpful to improve cold resistance.4.-4℃cold acclimation compared with 4℃for cold acclimation, enhance greater cold hardiness of grape roots.5.Off cold acclimation of the grape root, some cold resistance which low temperature exercise produced is disappeared.The results can be seen from the tests:The physiology of off low exercise group grape roots between low exercise group and control group at room temperature. This indicates that cold acclimation produced cold resistance partial disappeared.6.Grafting has some impact on cold hardiness of grape root, but the extent is not affected.Grafting has no significantly impact on cell membrane permeability and organizational structure,has a certain impact on water content, proline content, MDA content and soluble sugar content. Different scion grape roots have little differences on projects which has been mentioned above.The results of this test have some guidance on improving methods of grape cultivation and cold grape varieties breeding. |
