Epimedium is a member of the barberry family (Berberidaceae) of herbaceous perennials.At present, there are about 59 species in the world and 48 species in China, which Sichuan province has the most, about 22 species and is the main distribution area of the world. The Chinese Pharmacopoeia of 2010 edition has collected 4 species,they are E. sagittatum, E. pubescens, E.korearmm and E. brevicornu, Sichuan has 3 species of Epimediuml,including E.pubescens, E. brevicornu and E. sagittatum, the others are also used as the local medicinal plants in local area. The modern pharmacological results indicate that,the Epimedium are useful to kindney yang tonics,strengthen muscles and bones and rheumatism as a crude drug in traditional medicine to enhance kidney function, also they have a kind of estrogen, the effect of immunizing and lowering the blood pressure and BGC, because it can increase the blood flow rate and enhance blood generating, therefore they are widely used to treat climacteric syndrome, osteoporosis and heart blood vessel diseases such as CAD, angina pectoris and so on. They are also used to treat diseases as cancer for its function of resisting tumor, such as antioxidation, anti-aging, antitussive, antiasthmatic and so on. Epimedium species are used in traditional Asian medicine as a crude drug for curing several diseases,which also have tremendous potential for commercial development.Although the breeds of Epimedium are great variety and wide distribution, people's unconscious plucking leads the standard unmatched breeds to seak into the market and of course the quality and the efficacy are extremely unstable. As the Epimedium's capacity of sexual reproduction is low, with the addition of long-term excessive plucking and the ecological environment's destruction, they have already been in a miserable situation of resource lacking and even facing danger.This paper discussed the tissue culture, the content of Flavonoids and the inorganic elements of the Epimedium in Sichuan province, in order to supply theory basis for its rapid reproduction, high-grade germplasm resources screening and GAP base construction. The main results are as follows:1.The E pubescens as the research object, this thesis discussed the optimal culture condition of the Epimedium's callus including from three aspects:the choice of the explants, the proportion of the nutrient medium and the culture conditions including the sterilization time, the light and the temperature. The result is as follows:the optimal explants are petioles and spires, the explants' livability is the highest and the callus acquired is the most if they are cultivates in darkness at 21℃after having been sterilizated for 3 minutes; the optimal nutrient medium which has the strongest ability to induce callus is like this:2,4-D 3 mg/L+6-BA 1 mg/L+NAA 0.5 mg/L.2.The result of Flavone measurement indicates as follows:the composition of 8 species is different. The E. Chlorandrum and the E. rhimedium are the lowest, which are significantly lower than other species and the other 6 species do not have significant differences; according to the result of the different parts' composition measurement, the general Flavone in petiole are significantly lower than they in leaves and callus of the same specie; populations'composition presents extremely significant difference (P<0.01).The most composition of Flavone is in these three species:the E. Acuminatum of ShangLi, the E. pubescens of ZhouGongshan and the E. Davidii of BaoXing.3.To analyza the inorganic elements in the Epimedium in Sichuan and to provide data for the medicinal uses. Ca, Mg, Fe, Na, Zn, Cu and K were determined by flame atomic absorption spectroscopy, and Pb, Cr and Cd were determined by graphite furnace atomic absorption spectroscopy. The results are as follows:the quantity of Cr was in the most amounts and Cu was the least.the concentration of Ca, Mg, Zn and Na were the highest in E. acuminatum of emeishang,namely,63.76mg·g-1,14.99 mg·g-1,185.2μg·g-1 and 6.188 mg·g-1;the concentration of Fe, Cu were the highest in E. davidii of yanjing, namely,2.499 mg·g-1 and 57.20μg·g-1;the concentration of the harmful element Pb and Cd were high, and the concentration of Pb and Cd were the highest in E. chlorandrum of baoxing. Namely,1737μg·g-1 and 35.31 mg·g-1,the concentration of them are higher than that in The Chinese Pharmacopoeia. The principal components analysis of SPSS was applied to study the characteristic elements in Epimedium which were Ca, Na and Mg.4.Through the studies on the Flavonoids and Inorganic Elements of Epimedium in Sichuan, this research confirmed that the E. pubescens, E. acuminatum and E. flavum are the high quality species.Including, The E. pubescens, E. Acuminatum broad range of distribution, wide distribution, high yield, suitable for the production of large-scale cultivation, but only distributed in the E. Flavum, adaptability and yield relatively low, is not suitable for produceion use.
