Effect of Ghrelin and SCF on in vitro developmental competence of mouse embryos derived from in vivo (1-cell embryo), IVF and parthenogenesis was investigated, aimed at seeking for an effective factor that can improve developmental competence of embryos cultured in vitro.Two experiments were conducted and results were as follow:1.Effect of Ghrelin on the in vitro developmental competence of mouse pre-implantation embryos.(1) One-cell stage embryos recovered from mouse uterus were cultured in the presence of 0 ( control ), 0.1, 1, 10, and 100 ng/ml of human ghrelin, receptively. In each experiment, more than 10 embryos were assigned randomly in each group, and the experiment was repeated 7 times. No significant differences ( P>0.05 ) were detected for cleaved rate and the total number of blastocyst when cultured in HTF containing different concentration of Ghrelin compared with control group; the mourla rate and the blastocys rate decreased remarkably in HTF containing 0.1 ng/ml Ghrelin (P<0.05) compared with the control group, however, when more ghrelin was added to HTF medium, the embryos developmental competence recovered to some extent (P<0.05).(2) The same experiment was conducted wtih mouse IVF embryos as aforementioned. Blastocyst rate decreased remarkably when cultured in HTF containing 1ng/ml of Ghrelin (P<0.05) compared with control group, however, there's no significant differences for cleaved rate, morula rate and blastocyst cell number was detected between experimental groups and control group.(3) A total of 204 mouse parthenogenetic embryos were assigned randomly into 5 groups and cultured in HTF containing 0 (control), 0.1, 1, 10, and 100 ng/ml of human ghrelin, receptively, and results were similar to experiment conducted with in vivo fertilized embryos, i.e., the blastocyst rate decreased obvisously when cultured in HTF containing 0.1ng/ml l Ghrelin compared with control group, in spite the difference was not significant(P>0.05), and the blastocyst rate rised compared with 0.1ng/ml group when elevated ghrelin concentration.2. The effect of SCF on the in vitro developmental competence of mouse pre-implantation embryos.(1) we tested the effects of different concentrations of SCF on the in vitro developmental competence of mouse embryos derived from in vivo , results showed : there was not significantly difference for any embryos.(2) we tested the effects of different concentrations of SCF on the in vitro developmental competence of mouse embryos derived from IVF , results showed : the blastocyst rate and mourla rate increased significantly when cultured in KSOM containing 50ng/ml SCF (P<0.05) compared with control. (3) we tested the effects of different concentrations of SCF on the in vitrodevelopmental competence of mouse embryos derived from parthenogenesis , results showed that there was not significantly difference for any embryos.To sum up, Ghrelin had negative effects on the in vitro developmental competence of mouse pre-implantation embryos. In converse,SCF had positive effects on the in vitro developmental competence of mouse pre-implantation embryos.
