| Acremonium hansfordii is a kind of cosmopolitan epiphyte belonging to Moniliales, Hyphomycetes, adelomycete subphylum. In order to provide certain theoretical basis for production and application of Acremonium hansfordii preparation, the formula, preparation processing, toxicity test of emulsifiable suspensions and wettable powder of Acremonium hansfordii conidia and stockpile technology at normal temperature of conidia have been studied. Major conclusions are as follows:Processing of emulsifiable suspensions of Acremonium hansfordii conidiaFormulation for emulsifiable suspensions of Acremonium hansfordii Conidia was obtained through addition agent selection.The results indate that ginger oil was the optimal solvent naphtha, the mixture of tween-80 and OP-10 was the optimal emulsifier that its dosage was 0.3%, 0.1% sodium dodecyl sulfate was the wetting agent, 0.3% sodium carboxymethyl cellulose was the optimal stabilizing agent, 0.3% Humic acid was the optimal Uv protectant, streptomycin sulphate was used as antibiotics and its optimal concentration was 0.25%, glycol was optimal antifreeze and its proportion was 3%. Pour each addition agent into ginger oil according to theri proportion and stir well. Pour conidia power into the intermixture and stir well too. After the operation we got the emulsifiable suspensions of Ahy1conidia.Optimize the formula of wettable powder of Acremonium hansfordii conidiaIn order to enhance the practicality of wettable powder of Acremonium hansfordii conidia, the formula of wettable powder of Acremonium hansfordii conidia have be optimized by our lab. Screeded out the carrier was diatomite that had the least effect on the conidia activity, wetting agent was 1% sodium dodecyl sulfate, dispersant was 1.5% sodium lignosulphonate, agglutinant was 0.05% Xanthan, 0.1% zinc sulfate was used as promoting agents of spore germination, 0.1% Humic acid was the optimal Uv protectant, 0.25% antibiotics was streptomycin sulphate, and 0.3% sodium carboxymethyl cellulose was the optimal stabilizing agent.Toxicity test The drug-effect appraisement that emulsifiable suspensions and wettable powder of Acremonium hansfordii conidia indoor for Brevicoryne brassicae Linnaeus and Mythimna separata indicates the following results.The corrected mortality that two kinds of agents for testing insect was less in the first day, and it was increasing in the second day, then it was up to death peak in the third day showing insecticidal action obviously while the effect was obvious after 24h treated by Bacillus thuringiensis as control. The result that lethality of two kinds of agents for cabbage aphid was high compared with armyworm indicates that the effect of Acremonium hansfordii agents for Homoptera was high compared with Lepidoptera. The effect of mulsifiable suspensions of Acremonium hansfordii for aphid was high compared with the wettable powder. The corrected mortality was 88.4% when diluted 1000 times.Autologous physiology of conidiaAcremonium hansfordii conidia with different moisture content(1.73%, 6.31%, 21.4%)were stored at 4℃and 25℃separately. We have detected the active conidia rate, autologous nutrition including glucose and protein for 240d. The result indates that glucose content and protein content of treated conidia reduced slowly in the first three months. The livability of Acremonium hansfordii conidia with different moisture content at 4℃reduced slowly compared with 25℃. The active conidia rate reduced suddenly along with water content of conidia increasing. |
