China is the largest country that it producse apple and consumes apple,China is a decisive function in the world's apple industry. Marssonina mali is the one of apple early defoliation diseases.Until now,the prevention of Marssonina mali is mainly chemical prevention.The research of Marssonina mali's resistant mechanism and molecular marker assistant selection has very important value for the world's apple industry.The research at first establish the excised leaf tissue inoculation (ELTI) method about Marssonina mali,then evaluating the separation of the hybrid progeny's resistance,then according to the separation genetic analyses the Marssonina mali's resistant mechanism,at last,the selection of SSR molecular marker linked to the resistance of Marssonina mali,and hope to have good basis for molecular marker assistant selection in resistance breedingThe experiment started December, 2007 and ended April ,2009,using jonathan×golden delicious,adopted the ELTI method to study the identification of the resistance to Marssonina mali,identified resistant Marssonina Mali individuals and susceptical Marssonina Mali individuals ,and using SSR molecular marker to research resistant Marssonina Mali genes with BSA.The experiment has the following conclusions:1. The optimal method of the excised leaf tissue inoculation ablout Marssonina Mali was: twing the third and the forth joint's leaves of the underyearling branch,these leaves were consistent maturity.The most suitable temperature was 25℃,the most suitable concentration was 5×105 /ml,the most suitable deposited mode of the leaf is the foliar was upward with double-faced inoculated and foliar was upward with the foliar inoculated.And can observe the pathogenesis situation after the seventh day.2. Using the excised leaf tissue inoculation method to inoculate 1401 apple indviduals in Changli institute of pomology, Hebei Academy of Agricultural and Forestry Science Dongshahe apple nursery. The results revealed:the resistantce to Marssonina Maliof the hybrid progeny showed significant separation,by Chi-square showed that resistant indviduals and susceptible indviduals conformed to 3:1 separation rate,accounted for the inheritance of Marssonina Mali had major gene effect, major gene expressed the resistant to the susceptible is dominnat,the resistance was dominated by one pair or two pairs genes and minor-polygene. the genotypes of parents were Aa×Aa or Aabb×aaBb.3. Establish appropriate SSR reaction system for Marssonina Mali.using the 6-year-old jonathan×golden delicious seeding progeny as separate target,in 144 pairs SSR primers selected 17 paris which expressed polymorphism.And the primer pairs CH01h01 amplified a 300bp band in resistant and susceptible indviduals,it was the SSR molecular marker linked to Marssonina mali, its genetic distance is 10.1 cM.According to genetic linkage map,the marker linked to Marssonina mali located in LG8 of apple's the eighth linkage groups.
