| Flower bud of Chrsanthe memindieum, which has been used as a kind of drinks in Zhejiang Province for a long time, was chosen as experimental material to extract flavonoids, using the more advanced ultrasonic extraction, microwave extraction, supercritical CO2 extraction respectively. Compared with lixiviating method, we obtained the optimum extracting process of flavonoids from the flower bud of Chrsanthe memindieum. The antibacterial activity of extracts from the flower bud of Chrsanthe memindieum was also determined by bacteriostatic test, so as to providing theoretical basis and practical guiding for exploiting and utilizing the flower bud of Chrsanthe memindieum in natural preservative. The results as follows:1. With the lixiviating method and taking water as solvent, the optimum processing of extracting flavonoids was that: the ratio of material to liquid was1:100, the extracting time was 60min, the extracting temperature was 80℃. The yield of flavonoids was 10.36% under this conditions; with ultrasonic extraction method, the optimum processing of extracting flavonoids was that: the ratio material to liquid was1:120, the extracting temperature was 70℃, the extracting time was 40min. The yield of flavonoids was 16.96% under this conditions; With the microwave extraction method, the optimum processing of extracting flavonoids was that: the power of microwave was 800W, the extracting time was 9min, the ratio material to liquid was1:140. The yield of flavonoids was 18.83% under this condition; with the supercritical CO2 extraction method, the optimum processing of extracting flavonoids was that: the extracting pressure was 20 MPa, the ratio material to liquid was 1:15, extracting time was 2.5h, extracting temperature was 60℃. The yield of flavonoids was 0.82% under this condition. The microwave extraction method showed the best extraction effect of flavonoids among these methods.2. The bacteriostatic test of extracts from the flower bud of Chrsanthe memindieum showed that: the extracts had no antimicrobial activities on Aspergillus niger; it had certain antimicrobial activities on Saccharomyces carlsbergensis, Aspergillus flavus and Penicillium chrysogenum, but the bacteriostatic effect was not strong; it had strong antibacterial activities on Escherichia coli, Bacillus subtilis and Staphylococcus aureus. The results showed the extracts had better bacteriostasis on bacteria than fungi.3. The MIC values of Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Saccharomyces carlsbergensis, Aspergillus flavus, Penicillium chrysogenum are: 6.25%, 12.5%, 3.125%, 50%, 100%, 100%. The MBC values of Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Saccharomyces carlsbergensis are: 6.25%,25%,6.25%,100%。... |
