Certified reference materials are samples with well established properties used for the assessment of analytical methods.Reference materials have characteristic of accuracy,homogeniety and stability.Reference materials are valuable for method development and validation.In recent years,although there is great progress in modernization of quality of traditional chinese medicine many problems to be resolved urgently exist.So there is an important signification to enhance standard of traditional chinese medicine by explore traditional chinese medicine reference materials.This thesis sums up general of reference materials,research progress in traditional chinese medicine reference materials.A prepared method of quercetin and emodin is established using vacuum column chromatography and solvent washing. This method has some features that the prepared efficiency is high,the isolated effect is good and so on.On this basis,identify the candidate of reference materials by spectroscopy.The amount of quercetin prepared is 2.6924 g and emodin is 4.8 g. Additionally,HPLC methods are developed to characterise reference materials of flavonones in Ginkgo biloba L.and emodin.The chromatographic column is Agilent Zorbax SB-C18(4.6mm×250mm,5μm);mobile phase is MeOH:0.4%H3PO4= 50 :50 for quercetin and MeOH:0.4%H3PO4 = 55:45 for kaempferol and isorhamnetin;the column temperature is 30℃and the flow rate is 1 ml/min.The chromatographic column is Agilent Zorbax SB-C18(4.6mm×250mm,5μm ); mobile phase is MeCN:MeOH:0.1%H3PO4 = 30:40:30 for emodin;the column temperature is 30℃and the flow rate is 1 ml/min.The result of quercetin and emodin characterised is 99.42%and 99.57%by HPLC condition established.The result of characterisation of quercetin and emodin achieves requirement of reference material, repeatability of HPLC method is good.Homogeneity and stability of flavonones in Ginkgo biloba L.is well and related substances of emodin are in investigation.
