| Sika deer(Cervus Nippon) is a rare and precious wild animal in China,and it is at the first level of the national protection in wild animals.The antlers of Sika deer is a traditional medicine material. The economic efficiency of artificial rearing Sika deer is considerable.China is the earliest country of the world to feed Deer,but there is little science and technology to support the feeding and breeding Sika deer in a long term.Nowadays the primitive prectice still used in deer industry,so that the deer population quality serious degradation,performance decline,low quantity of antlers and weak resistance of disease.The artificial insemination technology of Sika deer is a useful way to improve deer population quality and increase the ultilization ratio of good bucks.Collecting and preserving semen is a key ring in the artificial insemination.We take northeast Sika deer in China as experiment object, comparing effects of different stabilization method on semen quality,comparing semen quality of different ages and checking semen quality of different times in the same breeding season;we also tried different scheme in sperm freezing and frozen-thawed,contrast effects of different scheme on semen quality and sperm living ability;we discussed the electroejaculation and sperm freezing preservation of Northeast sika deer under the condition of stable breeding,desiring to provide a theory and appliance foundation to Sika deer breeding.In the period from September 2005 to October 2007,the experiments were carried on at Guanjia deer farm,in the Xinbin Manchu Autonomous County of Liaoning Province,which included the semen collection,freezing of semen and so on.The results were as follows:1.Deep anesthesia stabilization is better than Mild anesthesia stabilization,the percentage of success is 100%.The voltage of ejaculation was(8.56±0.54) V.The volume of semen was (1.5±0.32) mL.The difference of semen quality between two methods was not significant. However,deep anesthesia stabilization is easier to operate so that more suitable to promote in deer farms.2.The semen quality of Sika deer is best in 6~7 years old phase;the Sperm motility of fresh semen is 0.82±0.076,the Sperm density is 10.06±1.52,the Abnormality percentage is (13.85±1.64)%,the Intact acrosome rate is(84.69±2.43)%;After frozen-thawed the Sperm motility is 0.42±0.063,the Abnormality percentage is(26.46±2.64)%,the Intact acrosome rate is (59.53±2.82)%.3.The Sperm Living ability of Sika deer is best in 6~7 years old phase;the Living time of fresh sperm under 4℃is(119±14) h,the living index is 24.5±2.4,and the Living time of fresh sperm under 37℃is(5.1±1.8) h,the living index is 1.84±0.45;the Living time of frozen-thawed sperm under 37℃is(2.5±0.8) h,the living index is 0.42±0.16.4.In the early of breeding season the result of electroejaculation and semen quality are both the best of all,throughout the breeding season in over time,the bucks physical decline,semen volume and sperm quality also declined.Therefore the semen of early breeding season is the best choice to freezing preservation and artificial insemination. |
