Wheat(Triticum aestivum) is the most important food crop in the world, and is the second important in our country except rice. It plays an important role in agriculture. Modern breeding leads to the narrow genetic diversity of breeding parents, so there is no further improvement in wheat breeding. The old wheat landrace as a germplasm is one of the important genetic variation sources. There are 13976 wheat landraces on record in our country. The landrace in our country carrying many specific genes created great influence on wheat breeding. In this experiment, seed storage protein, EST-SSR and identification of resistance to powdery mildew were analyzed in order to study the alleles diversity in Jiuquanjinbaoyin from different perspectives. The aim was to probe into genetic structure and point out its persistence and wide adaptability. The results were as follows:1. The seed storage protein in 97 inbred lines of Jiuquanjinbaoyin were analyzed by SDS-PAGE and A-PAGE. The results indicated that 94.85% of the lines were homogeneous at the Glu-1 loci, but there were much variation between different lines. 9 kinds of HMW-GS combination and 19 kinds of gliadin were found.2. Using EST-SSR to investigate Jiuquanjinbaoyin, we found that 13 out of 84 primer pairs were polymorphic. The function of these primers was analyzed. The results showed that 5 primers could improve the stress resistance ability, 1 primer might prevent self-pollination and the other 1 primer involved with seed storage protein.3. The results of powdery mildew resistance in Jiuquanjinbaoyin suggested that all of them were susceptible to XN strain while their resistance was different to other 4 stains (E05, E09, R15 and HN). The ratio between susceptible and resistant lines was 91:3, 88:6, 90:4 and 91:3. Moreover, there were 4 reaction types, which contained 2 kinds of high resistance (0; and 1). While in the control cultivar, Xiaoyan6 performed similar reaction to powdery mildew. Only 1 line was resistance to E09 and all other lines were susceptible to other 4 powdery mildew. The results showed the landrace were more heterogeneous than breeding cultivar in functional genes.
