Lentinus edodes (L. edodes)is a well known macrofungi with edible and medicinal value, its polysaccharides have a variety of biological and immunological activity, such as anti-tumor effects, stimulate the formation of interferon, strengthen host resistance to bacterial, fungal, viral or parasitic infections, enhance natural killer (NK), cytotoxic T lympjocutes (CTL) activity and so on. Therefore L. edodes have attracted great attention in medicine circles and have been becoming an attractive material as pharmaceutical products.In this paper: dry mushroom L. edodes were extracted with distilled water for three times, then the supernatant was concentrated and precipitated with ethanol to obtain the crude polysaccharide (designated C-LNT-1). C-LNT-2 was recovered by repeated frozen, thawed and precipitating with 95% ethanol from C-LNT-1. C-LNT-2 was deproteinated by a combination of proteinase and Sevag method and was further purified by Sepharose CL-6B gel filtration chromatography to get the homogeneity and low molecular weight lentinan(Mw=7040Da)(designated LMw-LNT)which was evaluated by HPLC. Gas chromatography (GC) analysis showed that LMw-LNT was composed of Glc, Gal, Man and Fuc in ratio of 9.35:3.53:2:1. Immunobiological activity mediated by LMw-LNT in ICR mice , which were immunized subcutaneously with ovalbumin (OVA) alone or with OVA dissolved in saline containing LMw-LNT, was evaluated in this paper. The results showed LMw-LNT significantly enhanced the ConA- or LPS-induced splenocyte proliferation; antibody titers (IgG, IgG1) against OVA in serum from mice immunized with OVA dissolved in LMw-LNT were increased significantly when compared with mice injected with OVA alone; while IgG2b significant differences were not observed between OVA-injected mice and OVA with LMw-LNT-immunized mice. The results suggest that LMw-LNT enhances immunologic activity of mice and may be consider as a potential adjuvant to improve the efficacy of vaccine.
