Sequence Analyses Of 3' Terminal And Polymerase Activity Module In L Gene Of Rabies Virus Wild Strains From 2006 To 2007

Rabies is an acute zoonotic infectious disease caused by Rabies virus (RABV). In recent years, it has been a horrible threat to our people's life safty in guangxi province. And molecular epidemiologic studies of RABV have been established in order to provide a better understanding of the evolution of this virus.In this study, the cDNA fragments of 3' terminal and polymerase activity module in L gene of 11 rabies virus wild strains in guangxi were cloned and sequenced. The nucleotide sequences of these cDNA fragments mentioned above and their deduced amino acid sequences were compared with that of rabies virus fixed strains, rabies-related viruse strains published previously together with the sequences of strains preserved in our laboratory. The results indicated that the homology of wild strains in guangxi was at a high level, the nucleotide and deduced amino acid sequence homologies of 3' termernal in L gene ranged from 85.6 to 100.0% and from 91.6 to 100.0%, and that of polymerase activity module ranged from 88.8 to 100.0% and from 97.5 to 100.0% respectively. Compared with some other rabies virus fixed strains and rabies-related viruse strains, the nucleotide and deduced amino acid sequence homologies of 3' termernal in L gene ranged form 67.5 to 96.8% and from 70.4 to 99.1%, and that of polymerase activity module ranged from 76.8 to 97.8% and from 94.0 to 100.0%. The analysis of the deduced amino acid sequences showed that the mutation positions of 3' terminal in L gene focused on position 2,17, 19, 48, 73, 74, 136, 161. And that of polymerase activity module in L gene focused on position 49, 63, 72, 108, 136, 145, 157. Furthermore, there is a core sequence "AQGDNQ" exists in position 117 to 122 which is strictly conservative.The nucleotide and deduced amino acid sequence sequence analyses of 3' terminal and polymerase activity module in L gene showed that the genetic relationship among all rabies virus wild strains in guangxi is close, and it is comparatively more different from the fixed rabies virus strains. And there is a great difference between strains in guangxi and rabies-related viruse strains. According to the phylogenetic analysis, the strains in this study can be grouped into the cladesâ… ,â…¡,â…¢which were divided already based on the strain sequences preserved in our laboratory, and that has consistence. GXQZD, GXHXB, GXLB, GXNND were grouped to cladeâ… which has GX08, GX09, GX014, GX091, GX195, GX260, GXHX, GXLA, GXWX, GXSL contained already; GXYZD, GXLA11, GXLC9, GXLCC, GXNN2, GXPL, GXPXD to cladesâ…¡with GX074, GX219, GX304, GXBM, GXPX contained. There is only GXN119 in cladeâ…¢without new one grouped to. It is coincident with theanlysis results based on the N,G genes of rabies virus.
