Isolated microspore culture technique has received much more attention because of its potential application prospect. In the Solanaceac,isolated microspore culture technique of tobacco has developed better. However,other Solanaceac crops such as eggplant,tomato,pepper which Isolated microspore induction rate is low and plant regeneration is very difficult.Three diploid cultivars of eggplan(tsolanum melongena L.)Z1,Z3 and Z5 were used in the present research. This experiment further study the heat shock signal expression and in vitro morphogenesis in eggplant (solanum melongena L.) microspore on the basis of prior research. The results of the present experiment showed as following:1. Observation on the microspore of heat shock 0d,2d,4d,6d,8d,the results showed that 36℃for 6d resulted in the highest swell ration and survival ration.2. Observation of microspore viability showed that viable and degraded microspores existed in a same microspore population. This phenomen fully explained the necessity that removed degraded microspores from fresh or treatment anther.3. In vitro culture the microspore of 36℃for 6d,the results showed that enlarged microspore had the highest survival ration and the underwent degeneration microspore died gradually. It is suggested that to obtain high level of microspore morphogenesis,the enlarged microspore population should be isolated from the degenerating ones for further in vitro culture.4.Heat shock was an important condition to change developmental pathway of microspore. The microspore by heat shock treatment produced callus finally. However the microspore by no heat shock treatment produced pollen tube. The production percentage of callus and the microspore pollen tube germination percentage were genotype-dependent.5. By semi-quantitative RT-PCR detection,we founded that 36℃for 6d HSP70 mRNA expression is higher than the no heat shock treatment.But there were differences between different varieties. Z1 was the highest. Second is Z5,Z3 was the lowest. This result compared with isolated-microspore morphogenesis (Z5>Z3>Z1) , we found that HSP70 mRNA expression and the potential of isolated-microspore morphogenesis is not in direct proportion.That is,the increase of mRNA is not equal to the increase of protein.6. Natural doubling rate of regenerated-plants which derived from microspore culture is high,about 50%. There was a very significant difference among leaves,flowers,stomata in haploid,diploid and tetraploid. There was no significant difference between plant height and plant width. Tetraploid plants which leaves became bigger and thicker. The fruit of haploid plants can't be found. Diploid plants which had lots of seeds and the seeds were very plump. The seeds of tetraploid plants were few and small. We also found the character segregation of regenerated plant generation. some characters tend to male parent and some tend to female parent.
